摘要
目的:探讨钛离子对人外周血单个核细胞(PBMC)表达骨代谢相关因子白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)和核因子-κB受体活化因子配体(RANKL)的影响。方法:分离培养人的PBMC,分为活化和未活化PBMC,与不同浓度的钛离子(0μmol/L、15μmol/L、50μmol/L、200μmol/L)作用18h,RT-PCR检测各组IL-1β、TNF-α、IL-6和RANKL的mRNA的表达。结果:15μmol/L、50μmol/L、200μmol/L浓度的钛离子均可明显使活化与未活化PBMC的IL-1β、TNF-α、IL-6和RANKL的表达增加(P<0.05),且活化PBMC的各因子表达随钛离子浓度升高而增加。结论:钛离子可促进PBMC的IL-1β、TNF-α、IL-6及RANKL表达增多,且活化后更为显著。
Objective:To explore the effect of titanium ions on the expression of bone metabolism related factors,namely interlenkin-1β(IL-1β),tumor necrosis-α(TNF-α),interlenkin-6(IL-6)and receptor activator for factor-KB ligand(RANKL)in peripheral blood mononuclear cell(PBMC)of human.Methods:PBMC were separated and cultured.PBMC were divided into pre-activated group and non pre-activated group,and were co-cultured with different concentration of titanium ions(0μmol/L,15μmol/L,50μmol/L,200μmol/L)for 18 h.RT PCR were performed to detect the expression of IL-1β,TNF-α,IL-6and RANKL mRNA.Results:Treated with concentrations of titanium irons with 15μmol/L,50μmol/L,200μmol/L in pre-activity and non pre-activated of PBMC,the expressions of IL-1β,TNF-α,IL-6and RANKL were increased significantly(P〈0.05).Higher was the concentration of titanium irons,higher the expression in pre-activated PBMC.Conclusion:Titanium ions may stimulate PBMC to secrete IL-1β,TNF-α,IL-6and RANKL,and increase expression in the pre-activated PBMC.
