Extraction of Se- polysaccharides From Wheat by Orthogonal Test

摘要 本实验以河北小麦为研究对象,采用水提醇沉法,并以超声波辅助提取小麦硒多糖。在单因素试验的基础上设计正交试验以优化试验方案。利用统计软件SPSS对正交试验结果进行方差分析可知,提取小麦硒多糖的最佳工艺条件为:浸提温度60℃、超声提取时间40min、料液比1∶60。根据最佳的工艺条件,测得每克小麦中多糖含量为0.00331g,且硒多糖的提取率为8.622%。通过原子荧光法测出每克小麦硒多糖硒含量为0.08210μg。 In this paper,the hebei wheat was used as an raw experimental material. Water- extraction and alcohol-precipitation method was the main way that was taken to get the Se- polysaccharide from the wheat with the help of the ultrasonic during the extraction period. Based on the single- factor test,a orthogonal test was designed to optimize this program. The orthogonal test results were analyzed with SPSS in the way of variance analysis. The results showed that the best condition to extract the Se- polysaccharide were as follow: water extraction temperature was 60℃,ultrasonic time was40 min and the solid- liquid ratio was 1: 60( w / v). Under this conditions,0. 00331 g polysaccharide could be get from each gram of this wheat and the extraction yield of the Se- polysaccharide was 8. 622%. The atomic fluorescence method was used to detect the content of Se in Se- polysaccharide and there was 0. 08210μg selenium per gram of Se- polysaccharide.
