慢性牙周炎患者龈下菌斑和龈上菌斑微生物群落分析 被引量:17

摘要 目的:分析慢性牙周炎患者龈下菌斑和龈上菌斑微生物群落结构。方法:30名慢性牙周炎患者,分别采集龈上菌斑和龈下菌斑,运用DGGE技术分析两组样本微生物群落结构,采用聚类分析(CA)、偏最小二乘法(PLS)找出两组间存在明显差异的条带,通过测序确定与区别条带最相近的微生物。结果:CA和PLS结果均显示慢性牙周炎患者龈下菌斑和龈上菌斑微生物群落结构存在显著差异;测序结果显示,福赛斯坦纳菌、牙龈卟啉单胞菌、齿垢密螺旋体、龈沟产线菌、卡氏卟啉单胞菌、普氏菌属、简明弯曲菌是龈下菌斑的相关优势菌,而索氏密螺旋体和有害新月形单胞菌是龈上菌斑的相关优势菌。结论:慢性牙周炎患者龈下菌斑和龈上菌斑微生物群落存在生物多样性及差异。 Objective: To analyze the microbial community of subgingival plaque and supragingival plaque in peri- odontitis individuals Method:30 periodontitis individuals with no systematic diseases were recruited.Their subgingival plaque and supragingival plaque samples were collected.The microbial community was analysed by denaturing gradient gel electrophoresis (DGGE).Bands with significant differences between the two groups were confirmed by cluster analysis (CA) and partial least squares (PLS), and then excised for further sequencing. Result: Microbial community diferences existed be- tween subgingival plaque and supragingival plaque in periodontitis subjects.DNA sequencing suggested that Tannerella forsythia, Porphyromonas gingivalis, Treponema denticola, Filifactor alocis, Porphyromonas catoniae, Prevotella sp., Campy- lobacter concisus associated with subgingival plaque,while Treponema socranskii, Selenomonas noxia were related with supragingival plaque. Conclusion:Diversity and diferenees of microbial community existed in subgingival plaque and supragingival plaque in periodontitis individuals.
