摘要
目的:建立一种人血清中卡马西平(CBZ)浓度的高效液相色谱(HPLC)检测方法。方法:采用HPLC法测定人血清中的CBZ浓度。结果:药物在0.5~20.00μg/m L浓度范围内线性良好,回归方程为y=0.3609x-0.0053(R2=0.9999),相对回收率为94.0%~99.0%,日内、日间RSD均小于4%(n=6)。血清样品在室温下放置24 h保持稳定,RSD为0.28%。结论:本方法线性范围大,灵敏度高,精密度好,回收率高,结果稳定可靠,可为临床给药方案提供参考。
Objective: To establish an HPLC method for the determining plasma concentration of Carbamazepine. Methods: Axxlaim PolarAdvantage II-CI8 chromatographic column (4.6 mm× 150 mm, 5 μm) was used; mobile phase: acetonitrile acetate buffer (pH=4.14)=50 : 50; internal standard: Phenobarbital. Results: Range of linearity of the standard curve of CBZ is 0.5-20.00 μg/mL. Regression equation of CBZ is y=0.3609x-0.0053(R2 =0.9999). The relative recovery is above 94 %, intra-day precision and inter-day precision RSD is less than 4 % (n=6). The research also discovered that the blood serum sample remains stable after it is laid aside under the room temperature for 24 hours, RSD is 0.28 % merely. Conclusion: A method for determining concentrations of CBZ in serum by HPLC was established, with a wide linear range, sensitive, accurate, high recovery rate, the results are stable and reliable. It can also be used for research on Pharmacokinetics clinically, provides scientific guidance to optimize individual drug dosage.
