摘要
本实验采用胃蛋白酶从鮟鱇(Lophius litulon)鱼皮中提取胶原蛋白,分析该酶溶性胶原蛋白的氨基酸组成和超微结构,并初步探讨体内外清除自由基机理。结果显示,该胶原蛋白的超微结构呈均匀网状分布,是良好的药物载体,体外清除自由基效果呈浓度依赖性,当浓度为10 mg/L时清除O^-2·和·OH能力最强,另外,体外实验显示其能显著抑制小鼠皮肤中丙二醛(MDA)生成,提高超氧化物歧化酶(SOD)、过氧化氢(CAT)、谷胱甘肽过氧化物酶(GSH-Px)和羟脯氨酸(Hyp)含量,当剂量为100 mg·kg^-1·d^-1时最好。初步研究表明,该胶原蛋白具有体内外清除自由基作用,其氨基酸组成、超微结构与清除自由基能力有关,提取的鮟鱇鱼皮酶溶性胶原蛋白清除自由基活力接近于之前提取的胶原蛋白肽。
The free radicals scavenging activity of pepsin soluble collagen from the Lophius litulon skin was preliminarily investigated for providing basic processing information. The result showed, this collagen can be a good drug carrier due to well-distributed network ultrastmcture, The collagen exhibited the inhibitory activity against O^-2· and·OH with a concentration-depend ent manner, and 10mg/L is better. It was confirmed in protecting skin-aging by significantly controlling the formation speed of malondialdehyde (MDA) and the reduction of the activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT), glutathione (GSH) and hydroxyproline (Hyp) in D-galactose induced aged skin, and 100 mg·kg^-1·d^-1 is better. We concluded, it is found its amino acid composition and ultrastructure correlated with the free radicals scavenging ability, and the free radical scavenging activity of this collagen was similar to that ofLophius litulon skin collagen peptide.
