摘要
目的探讨DNA结合抑制因子-1(Id-1)在子宫内膜异位症(EMs)在位及异位内膜中的表达及其与EMs细胞凋亡的关系。方法取60例EMs患者的在位内膜和异位内膜为实验组,30例非EMs患者的正常子宫内膜为对照组,采用免疫组化SP法检测Id-1蛋白的表达情况,TUNEL法测定细胞凋亡指数(AI),统计分析二者的相关性。结果无论是增殖期还是分泌期,正常子宫内膜、EMs在位及异位内膜Id-1蛋白表达阳性率依次升高,而AI值依次降低,差异有统计学意义(χ2=27.228,P=0.000;F=90.845,P=0.000)。随着临床期别增加,EMs在位及异位内膜Id-1表达水平增加,而AI值降低,差异有统计学意义(χ2=8.333,P=0.040;χ2=11.322,P=0.010;F=36.319,P=0.000;F=28.690,P=0.000)。正常子宫内膜和EMs在位内膜增殖期AI值低于分泌期,差异有统计学意义(t=-7.568,P=0.000;t=-3.435,P=0.001);而EMs异位内膜在增殖期和分泌期比较,AI值差异无统计学意义(t=0.200,P=0.842)。EMs在位和异位内膜Id-1蛋白表达水平和AI值呈明显负相关性(r=-0.307,P=0.017;r=-0.542,P=0.032)。结论 Id-1高表达在EMs发生发展中起重要作用,与抑制EMs细胞凋亡密切相关。
Objective To investigate the expression of inhibitors of DNA binding (Id) -1 in the eutopic and ectopic endometria of endometriosis(EMs) patients and its relationship with apoptosis. Methods The expressions of Id - 1 protein were detected in 60 EMs eutopic, 60 EMs ectopic and 30 normal endometria samples by immunohistochemical SP method. Meanwhile, the apoptosis index (AI) was detected in all the samples by the method of TUNEL and a statistic analysis was made to see their relation. Results The positive expression rate of Id - 1 protein increased from the normal endometria samples, EMs eutopic endometria samples, to EMs ectopic endometria samples ; while the AI value decreased successively. The difference was statistically significant ( χ^2 = 27. 228, P = 0. 000 ; F = 90. 845, P = 0. 000). As the clinic stage increased, the expression level of Id - 1 protein of EMs eutopic and ectopic endometria samples increased, whereas the AI value declined. The difference was statistical- ly significant ( χ^2 =8. 333, P = 0. 040 ; χ^2 = 11. 322, P = 0. 010 ; F = 36. 319, P = 0. 000 ; F = 28. 690, P = 0. 000) ; The AI value of normal endometria samples and EMs eutopic endometria samples in the proliferative phase was significantly lower than that of those in the secretory phase (t = - 7. 568, P = 0. 000 ; t = - 3. 435, P = 0. 001 ). However, the AI values showed no significant difference in the two phases of EMs ectopic endometria samples (t = 0. 200, P = 0. 842). The expression level and AI value of EMs eutopic and ectopic endometria samples presented clear negative relation ( r = - 0. 307, P = 0. 017 ; r = - 0. 542, P = 0. 032). Conclusion Overexpression of Id - 1 plays an important role in the pathogenesis and development of EMs, and it is closely related to the inhibition of EMs apoptosis.
