黄芪甲苷对脑缺血再灌注后脑组织MPO活性及TNF-α、IL-1β含量的影响 被引量:9

Effects of Astragaloside Ⅳ on the Activity of MPO and the Content of TNF-α,IL-1β after Cerebral Ischemia and Reperfusion

摘要 目的探讨黄芪甲苷对大鼠脑缺血再灌注后脑组织髓过氧化物酶(MPO)活性及肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)含量的影响。方法将36例只SD大鼠物随机分为假手术组、模型组、黄芪甲苷治疗组,每组12只。采用分光光度法及酶联免疫吸附法分别检测各组大鼠脑组织MPO活性及TNF-α、IL-1β含量。结果与假手术组相比,模型组大鼠脑组织MPO活性及TNF-α、IL-1β含量明显增高(P<0.01);与模型组相比,黄芪甲苷治疗组大鼠脑组织MPO活性及TNF-α、IL-1β含量明显减少(P<0.05)。结论黄芪甲苷可下调脑缺血再灌注后MPO活性及TNF-α、IL-1β含量,从而减轻炎症反应,发挥脑保护作用。 Objective To investigate the effects of astragaloside Ⅳ on the activity of myeloperoxidase( MPO) and content of tumor necrosis factor alpha( TNF-α),interleukin-1 beta( IL-1β) after cerebral ischemia and reperfusion. Methods Thirty-six Sprague-Dawley( SD) rats were randomly divided into sham-operated group,model group,astragaloside Ⅳ group. The methods of pectrophotometry and ELISA were respectively used to measure the activity of MPO and the content of TNF-α,IL-1β in brains tissue. Results Compared with those in sham-operated group,the activity of MPO and the content of TNF-α,IL-1β increased significantly in model group(P〈0. 01). Compared with those in model group,the activity of MPO and the content of TNF-α,IL-1β decreased significantly in astragaloside Ⅳ group(P〈0. 05). Conclusion Astragaloside Ⅳ can decrease the activity of MPO and the content of TNF-α,IL-1β after cerebral ischemia and reperfusion,and alleviate inflammation reaction which may be one of mechanisms of neuroprotective effects of astragaloside Ⅳ on cerebral ischemia and reperfusion injury.
