摘要 目的 :探讨TGFβ1和BMP2 对培养兔关节软骨细胞增殖和分化的影响 ,从而了解其在关节软骨损伤修复及骨关节炎 (OA)病理变化中的作用 ,方法 :采用光电镜、免疫细胞化学、液烁计数、流式细胞仪分析细胞周期等方法观察软骨细胞的超微结构和增殖、分化及其特异性的表达。结果 :TGFβ1显著增加了原代软骨细胞3 H TdR的掺入和使其进入S G2 /M期细胞比例增加。而BMP2 则对第 5代软骨细胞有明显的促增殖作用。结论 :TGFβ1能促进原代软骨细胞的增殖 ,但不能阻止其增殖能力随传代培养而减弱 ,而BMP2 则显著促进了第 5代软骨细胞的增殖 ,这可能与细胞的分化状态和细胞外基质 (ECM )。 Objective: To study the effects of transforming growth factor β1(TGFβ 1) and bone morphogenetic protein2 on proliferation and differentiation of rabbit articular chondrocytes(RAC).Methods:We observed the ultromicrostructure,proliferation and special phenotype of chondrocytes,using light and electronmicroscope,fluorimetery,liquid scintillometery,flow cytometry and immunocytochemisty and so on.Results:TGFβ 1 stimulates chondrocytes augment 3H TdR uptake and resulted in recruitment of RAC into S/G2M phase,but BMP 2 enhanced proliferation of subcultured.Conclusion:TGFβ 1 stimulates proliferation of chondrocytes in primary cultured in vitro,but the effects of TGFβ 1 on proliferation of RAC decreased with the number of in vitro monolayer subcultured.However,the relationship between the effects of BMP 2 on proliferation of subcultured chondrocytes and the status of cell differentiation,ECM and cytoskeleton might exist.
