鹅SLCLC25A6基因的克隆以及填饲后表达特征分析

摘要 SLC25A6(溶质载体家族25成员6),也称为ANT3,是交换ATP和ADP通过线粒体膜的载体。本研究以鹅总RNA为模板,通过3′-RACE和5′-RACE的方法,测序拼接得到1 485 bp的c DNA序列,含有897 bp完整的开放阅读框,共编码翻译298个氨基酸。经预测SLC25A6编码蛋白的分子量为32 792.1,等电点为9.73,为稳定亲水蛋白,且SLC25A6蛋白螺旋结构和折叠片聚集成球状。序列分析表明,鹅与红原鸡的核苷酸与氨基酸同源性比较高,系统进化表明其亲缘性较高。通过荧光定量PCR发现SLC25A6基因在多种组织中表达,且在肝脏和脂肪中的表达量显著高于肌肉组织;填饲处理后,胸肌中SLC25A6基因的表达量呈显著性下降,暗示SLC25A6基因主要在肝脏脂肪能量代谢过程中起重要作用,同时可能在肌肉的生长发育过程中起调控作用。本试验获得SLC25A6基因的完整序列并研究其表达规律,为进一步研究该基因的功能奠定基础。 SLC25A6 (solute carrier family 25,member 6),is also known as ANT3 which exchanges ATP and ADP through mitochondrial membrane. In this study, total RNA of the Langdee goose was used as a template to obtain 2 fragments of SLA25A6 genes by RACE methods,a full-length of 1 485 bp SLC25A6 eDNA was acquired which contains a 897 bp long complete open reading frame,encode of a 298-amino acid polypeptide with a calculated relative molecular mass of 32 792.1 ku and pI of 9.73. The SLC25A6 was a kind of hydrophilie and stable protein, and SLC25A6 spiral structure and protein folding together into a ball. Sequence analysis showed that the Anser anser and Callus gallus nucleotide and amino acids homology were relatively high. Phylogenetic tree analysis showed the high homology among them. The real time fluorescence quantitative PCR detection of the expression SLC25A6 gene in breast muscle significantly increased after overfeeding treatment. SLC25A6 expressed in multiple tissues, and SLC25A6 genes expression in liver and fat were significantly higher than muscle tissue. After over feeding,SLC25A6 genes expression in chest muscle was significantly decreased, suggesting SLC25A6 genes mainly play an important role in the process of energy metabolism in fat and liver,meanwhile might play a key regulatory role in muscle growth. The experiment has obtained a complete SLC25A6 gene cDNA sequence and studied its expressional pattern,it lay the foundation of the function of the gene in future study.
