摘要 目的:通过检测胃癌组织内肿瘤相关性巨噬细胞(TAMs)中MiR-146a的表达,探讨MiR-146a对TAMs的影响。方法:采用RT-PCR法分别检测BALB/c小鼠BGC-823移植瘤和腹腔中TAMs、胃癌患者肿瘤组织及外周血单核细胞中MiR-146a的表达量;转染MiR-146amimics的巨噬细胞与BGC-823细胞混合接种于小鼠,观察TAMs中MiR-146a的表达对肿瘤生长的影响。结果:1)小鼠BGC-823移植瘤TAMs中MiR-146a表达量显著降低,MiR-146amimics能明显抑制小鼠BGC-823移植瘤的生长。2)人胃癌组织TAMs中MiR-146a表达水平越低,肿瘤分化程度越差,肿瘤体积越大;与年龄、性别、肿瘤浸润深度、淋巴结转移及远处转移无关(P>0.05)。结论:MiR-146a作为抑癌基因调节TAMs的功能,抑制胃癌的发生发展。 Objective: This study aimed to detect the expression of MiR-146a in tumor asso- ciated macrophages (TAMs), and explore the effect of MiR-146a on TAMs. Methods: the expres- sion of MiR-146a in TAMs of BGC-823 xenografted tumor and peritoneal macrophage in BALB/c mouse, TAMs of human gastric cancer and peripheral blood mononuclear cells were detected by RT-PCR method. The BALB/c mouse were inoculated with BGC-823 cells and macrophage with MiR-146a mimics transfection to determine the effect of the expression of MiR-146a on the volume and weight of tumor. Results: The expression of MiR-146a in TAMs of BGC-823 xenografted tu- mor was significantly reduced (P〈0,05), and MiR-146a mimics in TAMs significantly inhibited the growth of BGC-823 xenografted tumor (P〈0.05). The expression of MiR-146a in human gastric cancer tissues(P〈0.05) was significantly reduced, there was a negative correlation between expres- sion of MiR-146a and tumor size. and the expression of MiR-146a had no correlation with age, gen- der, tumor invasion, lymph node metastasis and distant metastasis (P〉0.05). Conclusion: These results suggested that MiR-146a in TAMs play a role in cancer suppressor gene regulating function of TAMs to inhibit the development of tumor.
