摘要
目的:探讨S100A4基因和STAT3基因在甲状腺乳头状癌组织中的表达及意义。方法:83例甲状腺乳头癌患者术中留取肿瘤及对侧正常甲状腺组织,利用实时荧光定量PCR技术对S100A4mRNA和STAT3mRNA的表达进行检测。结果:S100A4mRNA和STAT3mRNA在甲状腺乳头状癌组织中相对表达量均高于正常甲状腺组织(P<0.05);甲状腺乳头状癌组织中S100A4mRNA和STAT3mRNA相对表达量均与年龄、性别、肿瘤大小、分化程度无关(P>0.05),而与包膜浸润、TNM分期和淋巴结转移有关(P<0.05);甲状腺乳头状癌组织中S100A4mRNA相对表达量与STAT3mRNA相对表达量呈正相关(r=0.626,P<0.001)。结论:S100A4和STAT3基因在甲状腺乳头状癌组织中呈高表达,可能是JAK1-STAT3信号通路通过激活S100A4蛋白表达而实现对肿瘤细胞侵袭、转移等生物学过程的调控。
Objective: To investigate the expressions and significances of S100A4 gene and STAT3 gene in papillary thyroid carcinoma. Methods'. The tumor tissues and contralateral normal tissues of 83 cases of papillary thyroid carcinoma patients undergoing elective surgical resection were collected. The expressions of S100A4 mRNA and STAT3 mRNA in papillary thyroid carcinoma were detected by using real-time quantitative PCR. Results: The relative expression levels of S100A4 mRNA and STAT3 mRNA in papillary thyroid carcinoma were higher than normal thyroid tis- sue, the differences were statistically significant (P〈0.05). The relative expression levels of S100A4 mRNA and STAT3 mRNA in Papillary thyroid carcinoma were Not related with age, gender, tumor size. differentiation degree (P〉0.05), but were related with capsular invasion, TNM stage and lymph node metastasis (P〈0.05). Pearson correlation analysis showed the relative expression levels of S100A4 mRNA were positive related with the relative expression levels of STAT3 mRNA(r=0.626, P〈0.001 ). Conclusions: The expressions of S100A4 and STAT3 gene in papillary thyroid carcinoma showed high. It might be though the JAK1-STAT3 signaling pathway to activate the expression of S100A4 protein to regulate the biological processes of cell invasion, metastasis, etc.
