中药丹参及复方丹参片17种有效成分抗氧化作用的均匀设计-高通量筛选研究 被引量:1

The Application of Uniform Design-High Throughput Screening on Antioxidant Research of 17 Components Radix Salviae Miltiorrhizae and Compound Danshen Tablets

摘要 目的:在前期研究的基础上,以丹参及复方制剂复方丹参片有效成分组合活性评价为切人点,通过已经建立的多种抗氧化微孔板实验方法,探讨多因素、多水平均匀设计-高通量筛选(uniform design-high throughput screening,UD-HTS)中药研究的新模式。方法:通过SpcctraMax M5连续光谱酶标测试仪选择具有药理活性的丹参有效成分11种(其中脂溶性成分4种,水溶性成分7种)及复方丹参片有效成分6种,通过均匀设计,进行各种成分及不同浓度之间相互配伍组合,以抗2,2-二苯基-1-苦肼基自由基(2,2-diphenyl-1-picrylhydrazyl,DPPH)氧化活性、邻二氮菲-Fe^(2+)氧化法、还原能力测定及抗脂质过氧化法,评价各种有效成分组合对药效的影响。结果:经过初筛和复筛,初步得到了丹参最佳水溶性配伍、水溶性+脂溶性配伍及优化组合配伍的组合样品。结论:丹参各有效成分不同浓度均匀设计配伍,结合抗氧化高通量筛选技术可适用于传统多因素、多水平组合特点的大规模药效筛选研究的新方法。 Objective:On the basis of the previous studies,and further evaluate and research the bioaetive components group of Radix Salviae Miltiorrhizae (RXM) and compound Danshen tablets by multifactor and multilevel uniform design-high throughput screening method,to find a new pattern to research traditional Chinese medicine. Methods:By using SpectraMax M5 continuous spectrum enzyme sign reflectoscope reflector, 11 of bioactive components which been confirmed with pharmacological activities of RXM and compound Danshen tablets (4 of water-solubility and 7 of liposolubility) and compound Danshen tablets (6) were recombined to be a new compatible group through uniform design. And the effects of the anti-oxidant microplate quantification experiment models were detected by anti-DPPH effect, eliminating ratio of hydroxy free radical, reduction ability determination, and lipid peroxidation were established. Results:After high-throughout screening and rescreening,better watersolubility,better water-solubility add liposolubility,and optimal combination compatibility of RXM were obtained. Conclusion :The results indicated that effective component of RXM compatibility of different concentrations of uniform design, and with uniform design-high throughput screening of anti-oxidant models of microplate quantification referred to research bioactive component of compound recipe of traditional Chinese medicine.
