摘要
目的探讨围绝经期女性雌二醇水平对丝裂原活化蛋白激酶磷酸酶-1(MKP-1)代谢通路的调控与高发的抑郁症的关系。方法根据汉密尔顿抑郁量表(HAMD-17)评分及ICD-10"抑郁发作"的诊断标准入选196例研究对象并采集外周血标本。采用电化学发光法检测雌二醇(E_2),ELISA双抗体夹心法检测MKP-1蛋白水平,比较各组之间的差异及其相关性。结果围绝经期抑郁症组与其他三组比较,E_2水平降低(Z值分别为-1.887、-6.561、-7.563,均P<0.05),MKP-1水平升高(P<0.05),HAMD评分和E_2分级呈负相关关系(R=-0.227,P=0.033);HAMD与MKP-1显示正相关(R=0.287,P=0.000);E_2分级与MKP-1显示负相关(R=-0.264,P=0.000)。结论围绝经期雌激素水平的降低引发MKP-1水平升高,可能导致抑郁症的高发。
[ Abatract] Objective To study the regulation of estrogen level on mitogen-activated protein kinase phosphatase-1 ( MKP-1 ) and its relationship with high incidence of depression among perimenopausal women.Methods In accordance with the diagnostic criteria of depressive episode of ICD and Hamilton Depression ( HAMD-17 ) rating scale, 196 women were recruited and their blood samples were collected.Peripheral blood estradiol ( E2 ) was detected by electrochemiluminescence assay, and ELISA was used to detect MKP-1 protein level.The difference and relevance among different groups were compared.Results Compared to the other three groups , E2 level in the perimenopausal depression group decreased(Z value was -1.887, -6.561 and -7.563, respectively, all P〈0.05),but MKP-1 level increased (P〈0.05).HAMD scores were negatively correlated with E2 grading(R=-0.227,P=0.033)but positively correlated with MKP-1 levels(R=0.287,P=0.000).E2 levels were negatively correlated with MKP-1 levels (R =-0.264,P=0.000).Conclusion The decrease of estrogen level of perimenopausal women causes the increase of MKP-1 level, which probably leads to the high incidence of perimenopausal depression.
