HOXA10和HOXA11在子宫内膜异位症组织中的表达及相关性分析 被引量:4

摘要 目的探讨HOXA10和HOXA11基因在子宫内膜异位症患者异位内膜和在位内膜中的表达及二者之间的相关性。方法采用半定量RT-PCR反应和免疫组化SP法检测异位内膜60份、在位内膜25份和正常内膜35份中HOXA10和HOXA11基因的m RNA和蛋白表达情况。结果异位内膜中HOXA10m RNA表达明显低于在位组和对照组,差异极显著(P<0.01);异位组HOXA11m RNA表达显著低于在位组和对照组(P<0.05);异位组HOXA10蛋白表达显著低于在位组和对照组(P<0.05);异位组HOXA11蛋白表达显著低于对照组(P<0.05);而在位组HOXA11的表达也低于对照组,显著性差异(P<0.05),同异位组差异不显著。结论 HOXA10与HOXA11在内异症患者的子宫内膜中均低表达,而且呈正相关。HOXA10在子宫内膜的重塑中发挥作用,HOXA11会调节子宫内膜细胞外基质成分的改变。二者在内异症的发生发展起着相互协同的作用。 Objective To investigate the expression of HOXA10 and HOXA11 gene in the ectopic and of patients with endometriosis and and the correlation between them. Methods Semi-quantitative RT-PCR reaction and immunohistochemistry SP method were used to detect the m RNA and protein expression of HOXA10 and HOXA11 in 60 ectopic, 25 eutopic and 35 normal endometrium. Results The HOXA10 m RNA expression in ectopic endometrium was significantly lower than the eutopic and control group and the difference was significant(P 〈0.01). The HOXA11 m RNA expression in ectopic group was significantly lower than the eutopic and control group(P 〈0.05). The protein expression of HOXA10 in ectopic group was significantly lower than the eutopicand control group(P 〈0.05). The protein expression of HOXA11 in ectopic group was significantly lower than control group(P 〈0.05). However, there is no significant difference between the ectopic and eutopic group. Conclusion The HOXA10 and HOXA11 genes showed low expression in the endometrium of patients with endometriosis and were positively correlated. HOXA10 played a role in endometrium remodeling and HOXA11 changed endometrial extracellular matrix components. They had synergistic effect in the occurrence and development of endometriosis.
