摘要
从9种大孔树脂中筛选出吸附性能和解析性能良好的XAD-16型大孔树脂。采用高效液相色谱法评价大孔树脂吸附纯化的效果。通过单因素试验确定大孔树脂纯化恩拉霉素工艺。结果表明:当上样液pH为7.0,以2 BV/h的流速上样3BV供试液,然后用pH 3.0的体积分数为70%的甲醇溶液,以3 BV/h的流速洗脱5BV。在此条件下,纯化后纯度达85%,该样品较纯化前提高了1.83倍,且回收率达92.8%。
Purification process of enlamycin through macroporous resin was studied. XAD-16 macroporous resin showed the best adsorption and desorption capacity among nine kinds of macroporous resins. The concentrations of en- ramycin were evaluated with HPLC method. The detailed process conditions were as follows: the sample load was 3 BV, pH was adjusted to 7.0, sample loading rate was 2 BV/h, the eluent was 70% methyl alcohol(v/v) , pH was adjusted to 3.0, the dosage was 5 BV, with elution rate of 3 BV/h. After treatment with macroporous resin XAD-16, the purity of enramycins reached 85% , which was 1.83 folds higher than that by coarse extraction. The recovery rate was 93.2%.
