摘要
目的观察当归四逆汤对实验性心肌缺血模型大鼠血清一氧化氮(NO)及心肌一氧化氮合酶(eNOS)活性的影响,为临床应用本方防治冠心病提供实验依据。方法将50只雄性Wistar大鼠随机分为空白组、模型组、当归四逆汤低剂量组[5 g生药/(kg·d)]、当归四逆汤高剂量组[10 g生药/(kg·d)]和复方丹参滴丸组[0.081 g/(kg·d)],每组10只。各给药组均按30 m L/kg灌胃给予相应药物,空白组和模型组灌胃同体积蒸馏水,1次/d,连续5 d。末次灌胃后1 h,除空白组腹腔注射生理盐水外,其余组均腹腔注射垂体后叶素30 IU/kg。注射后1 h处死大鼠,测定各组血清一氧化氮(NO)水平,并取心尖部心肌组织采用免疫组化法检测内皮型一氧化氮合酶(eNOS)活性。结果模型组血清NO水平明显低于空白组(P<0.05),各给药组血清NO水平显著高于模型组(P均<0.05),当归四逆汤低剂量组和高剂量组比较差异无统计学意义(P>0.05)。光镜下显示,模型组eNOS活性弱于空白组,而各给药组eNOS活性均强于模型组。结论当归四逆汤可提高心肌缺血模型大鼠心肌组织eNOS活性,促进内源性NO生成和释放,从而发挥抗垂体后叶素所致大鼠心肌缺血作用。
Objective It is to observe the effect of Dangguisini decoction on the expression of serum NO and eNOS in myo -cardial tissue in experimental myocardial ischemia rat model , and to provide experimental evidence for clinical application of coronary heart disease .Methods 50 male Wistar rats were randomly divided into blank group , model group , low dosage of Danggi sini decoction group [5 g drug/(kg.d)], high dosage of Danggi sini decoction group [10 g drug/(kg· d)] and com-pound Danshen pill group [0.081 g/(kg· d)], each group had 10 rats.Every treatment group was given respective drug 30 mL/kg by intragastric infusion , blank group and model group were given the same volume of normal saline , one time per day and treated for 5 days.In one hour after the last time of Intragastric infusion , all the groups except blank group were given pi-tuitary oxytocin30 IU/kg by intraperitoneal injection .In one hour after injection , the rats were killed and their serum NO level and eNOS activity in apical myocardial tissue were detected by immunohisto chemical method .Results The serum level of NO in model group was lower than that in blank group (P〈0.05), while that in every treatment group were higher than that in model group (P all〈0.05), there was no significant difference between low dosage and high dosage groups of Dangguisini de -coction (P〉0.05).Light microscopy showed that the eNOS activity of the model group was weaker than that of the control group , while the activity of every treatment group were higher than that in model group .Conclusion Dangguisini decoction can enhance the activity of eNOS and promote the generation and release of endogenous NO , thus to play the protecting effect in the myocardial ischemia induced by pituitrin in rats .
