细胞因子诱导的杀伤细胞对鼠膀胱癌的抑制作用及T淋巴细胞亚群的影响

摘要 目的探讨细胞因子诱导的杀伤细胞(CIK)对膀胱癌小鼠的抑制作用及T淋巴细胞亚群的影响。方法取6周龄雌性无胸腺裸小鼠60只,随机分为治疗组、模型组及正常对照组,每组20只。治疗组建立膀胱癌原位荷瘤模型,并给予CIK处理,模型组则仅进行模型建立,给予同样剂量的0.9%氯化钠溶液输注,正常对照组不作任何处理,治疗上予同样0.9%氯化钠溶液处理。3组小鼠均于完成治疗后7 d处死,解剖小鼠,测量肿瘤的体积及质量,治疗过程中检测T淋巴细胞亚群CD3+、CD4+、CD8+、CD4+/CD8+的变化。结果治疗组与模型组比较,肿瘤体积[(145.34±46.32)mm3 vs(552.33±133.32)mm3]、质量[(0.18±0.23)g vs(0.76±0.21)g]差异有统计学意义(P<0.05)。CIK治疗使小鼠外周血CD4+及CD8+细胞增多,治疗组与模型组比较,CD8+差异有统计学意义(29.34±4.65 vs 15.52±0.12,P<0.05)。结论 CIK对膀胱癌起到抑制的作用,对膀胱癌有良好的疗效。 Objective To investigate the inhibition effects of cytokine-induced killer cells(CIK) on bladder cancer in mice and the influence on T lymphocyte subgroups. Methods Sixty 6-week-old female athymic nude mice were randomly divided into treatment group, model group and normal control group, n = 20 in each group. The treatment group was established as tumor- bearing orthotopie bladder cancer model and performed CIK treatment. Model group was established model only, and given the same dose of 0.9 % saline infusion. Normal control group was given no treatment but the same physiological saline. The volume and quality of tumor in three groups were measured after sacrificed the mice on 7 days treatment, and changes of CD3^+, CD4^+, CD8^+, CD4^+/CD8^+ in T lymphocyte subgroups during treatment were detected. Results Compared the results of treatment group with that of model group, the volume[(145.34 ± 46.32) mm^3 vs (552.33 ± 133.32) mm^3] and weight[(0.18 ± 0.23) g vs (0.76 ±0.21) g] of treatment group were reduced, and the differences were statistically significant(P 〈 0.05). The CD4^+ and CD8^+ cells of mice peripheral blood were increased after CIK treatment, compared with that of model group, and difference of CD8^+ was statistically significant (29.34 ±4.65 vs 15.52 ± 0.12; P〈 0.05). Conclusion It is demonstrated that CIK has inhibitory effect on bladder cancer with good therapeutic outcome.
