摘要
目的研究四君子汤复方多糖对荷瘤小鼠免疫功能的影响。方法建立小鼠S180实体瘤模型,分别给予低、中、高剂量四君子复方多糖,测定各组小鼠肿瘤生长抑制率、脾脏指数和胸腺指数;采用MTT法测定荷瘤小鼠脾淋巴细胞增殖作用;流式细胞技术(FACS)检测脾细胞表面抗原CD4、CD8的表达;酶联免疫吸附实验法(ELISE)测定小鼠血清白细胞介素-2(IL-2)γ-干扰素(IFN-γ)含量。结果中、高剂量的四君子汤复方多糖对S180肉瘤的抑制率分别为20.26%和23.28%,与模型组比较有显著性差异(P<0.05);中、高剂量的四君子汤复方多糖可以显著提高荷瘤小鼠的脾脏指数和胸腺指数;模型对照组小鼠CD4/CD8比值较正常小鼠明显下降,而四君子汤复方多糖可以显著提高荷瘤小鼠CD4/CD8比值;四君子汤复方多糖实验组小鼠血清IL-2和IFN-γ的含量都显著高于模型组小鼠(P<0.05)。结论四君子汤复方多糖具有显著的免疫增强活性,可以改善荷瘤小鼠的免疫损伤状况,发挥一定的抑瘤作用。
Obective To study immunological Effect of total polysaccharides from Sijunzi decoction (SJP) in tumor-bearing mice. Methods S180 tumor-bearing mice models were established and were given three different doses of SJP. The tumor growth inhibitory rate, spleen index and thymus index were calculated. MTT assay was used to observe the effect of SJP on spleen lymphocyes in vitro separated from tumor-bearing mice. The expression of surface antigens CD4 and CD8 in the spleen were detected by flow cytometry technique. The contents of IL-2 and IFN-γ were determined by means of ELISA. Results The tumor inhibition rates of medium and high-dose SJP were 20.26% and 23.28%, significantly compared with the model groups. Spleen index and thymus index were significantly inereased in medium and high-dose SJP groups.The ratio of CD4/CD8 in model group was lower than in normal group, SJP could up-regulated markedly CD4/CD8 value in tumor-bearing mice. The levels of IL-2 and IFN-γ in serum from SJP-treated mice, increased significantly compared to model control group (P〈 0.05 ) . Conclusion SJP had strong immunostimulating activity and could improve immunological damage in tumor-bearing mice.
