摘要
建立高分离度快速液相色谱RRLC法同时测定龟苓膏中绿原酸、芦丁的方法。采用采用ZORBAX SB-C18柱(150 mm×4.6 mm,1.8μm),乙腈-0.4%磷酸水溶液为流动相的梯度洗脱模式分离各测定组分,采用可变波长的方式测定相应组分的峰面积并计算样品含量。绿原酸和芦丁分别在0.009 42μg^0.471μg(r=0.999 9)和0.000 85μg^0.044 25μg(r=0.999 8)范围内具有良好的线性关系;且平均加样回收率分别为101.77%(RSD=1.2%,n=6)和99.84%(RSD=1.4%,n=6);重复性试验,2个成分含量的RSD均小于2.0%(n=6)。
To establish a RRLC method of simultaneously determining the content of chlorogenic acid and rutin in Guiling Paste. Samples were separated by ZORBAX SB-C18column(150 mm×4.6 mm,1.8 μm). Acetonitrile-0.4 % phosphoric acid was mobile phase with both a gradient elution mode and a variable detecting wavelength in order to determine the concentration of the respective components simultaneously. Chlorogenic Acid and Rutin all had good linearity in the ranges of 0.009 42 μg-0.471 μg(r=0.999 9) and 0.000 85μg-0.044 25 μg(r =0.999 8), correspondingly. The average recoveries were 101.77 %(RSD =1.2 %, n =6); and 99.84 %(RSD=1.4 %, n=6), respectively. Repeatability experiments presented that RSD values of their contents were less than 2.0 %.
