补肾健脾方药对尾部悬吊大鼠骨组织形态学影响的研究 被引量:4

Effects of Tonifying Kidney and Invigorating Spleen Prescription on the Bone Histomorphometry in Rat Tail Suspension Model

摘要 目的:观察补肾、健脾和补肾健脾三方对尾部悬吊大鼠骨形态计量学的影响。方法:将大鼠按随机数字表法分为正常组、悬吊组、补肾组、健脾组和补肾健脾组5组。除正常组外,其余各组大鼠采用头低位-30°尾部悬吊法模拟失重,连续悬吊21d。补肾组、健脾组、补肾健脾组每天给予对应方药灌胃,其余各组给予等体积生理盐水灌胃。实验21 d后,ELISA法检测血清骨钙素含量、双能X射线骨密度分析仪检测骨密度、股骨硝酸银染色、胫骨HE染色观察骨组织形态。结果:与正常组比较,悬吊组血清骨钙素、股骨骨密度、股骨骨小梁面积百分率、厚度、数量均明显降低,骨小梁分离度显著增高,胫骨骨小梁明显稀疏;与悬吊组比较,补肾组、健脾组和补肾健脾组各项指标明显改善,且以补肾健脾组改善较为明显。结论:补肾、健脾和补肾健脾三方均对模拟失重引起的骨丢失具有防护作用,能有效改善骨形态计量学参数,以补肾健脾方药作用为优。 Objective:To investigate the effect of tonifying kidney prescription, invigorating spleen and tonifying kidney and invigorating spleen prescription on the bone histomorphometry of tail-suspended rat. Methods: 50 male Wistar rats were randomly divided into five groups: the normal group, the suspension group, the tonifying kidney group, the invigorating spleen group and tonifying kidney and invigorating spleen group. Except the normal group rats, The remaining rats were suspended by head down tilt -30o for 21 days. Rats in the tonifying kidney group, the invigorating spleen group and tonifying kidney and invigorating spleen group were orally administrated with three decoctions and other groups with equivalent saline. Experiment 21 days, Osteocalcine was detected by ELISA method; Bone mineral density was measured using dual energy X ray absorptiometry, Through Silver nitrate staining of femur, HE staining of Tibial bone to observe the morphology of Bone tissue. Result: Compared with the normal group, The suspension group serum osteocalcin was significantly lower, femur bone mineral density was significantly lower, the trabecular bone area, trabecular thickness and trabecular number of femur bone were significantly lower, Trabecular separation significantly higher, trabecular obviously sparse of Tibial bone; Compared with the suspension group, the indexes of the tonifying kidney group, the invigorating spleen group and tonifying kidney and invigorating spleen group has improved significantly, and the tonifying kidney and invigorating spleen group improved obviously. Conclusion: Tonifying kidney prescription, invigorating spleen and tonifying kidney and invigorating spleen have protective effect on the simulated weightlessness induced bone loss, can effectively improve the bone histomorphometry parameters, and the effecs of the tonifying kidney and invigorating spleen prescrpition was obvious.
