岩大戟内酯B诱导乳腺癌细胞凋亡的实验研究 被引量:6

摘要 目的探讨岩大戟内酯B对乳腺癌细胞MDA-MB-435S凋亡的影响。方法将不同浓度的岩大戟内酯B作用于MDA-MB-435S细胞,采用流式细胞仪检测其诱导的细胞凋亡情况;电镜观察细胞形态。结果随着岩大戟内酯B浓度增加,细胞凋亡率明显增加;电镜显示药物处理后细胞出现核固缩,细胞器空泡变性,出现凋亡小体,甚至坏死。结论岩大戟内酯B可诱导乳腺癌细胞凋亡,为临床开发新药提供理论依据。 Objective To investigate the effects of Jolkinolide B induced apoptosis of breast cancer MDA-MB-435 S cells. Methods Apoptosis was detects by AnnexinⅤ-FITC / PI staining by flow Cytometry. Cell morphology was observed by electronic microscopy. Results Treated with Jolkinolide B,the rates of cell apoptosis were increased significantly( P〈0. 01). Many nuclei had become pyknotic by electronic microscopy,organelles empty bubble degeneration, appeared apoptotic body, even necrosis after dosing the different concentration of drug. Conclusions Jolkinolide B can significantly induced apoptosis of MDA-MB-435 S breast cancer cell. It provides the theoretical basis for a new drug with the development of clinical therapy.
