摘要
目的:研究外源性Apelin-13对急性心肌梗死大鼠心肌细胞保护作用。方法:通过结扎左冠状动脉前降支方法建立急性心肌梗死大鼠模型,存活大鼠分为心梗组(AMI组)及Apelin-13治疗组,另设假手术组(Sham组)及假手术Apelin-13治疗组(Sham+Apelin-13组)。治疗14d后,取心脏组织,TTC/Evans Blue染色检测心肌梗死面积;HE染色检测心脏组织病理变化;TUNEL法检测心肌组织细胞凋亡阳性率;Western blot法检测凋亡蛋白Bcl-2、Bax、Cleaved-Caspase-3的表达。结果:与急性心肌梗死模型组相比,Apelin-13治疗后,大鼠心脏组织梗死率显著减小,心肌组织炎性细胞富集减少,凋亡心肌细胞数减少,促凋亡蛋白Bax、cleaved-Caspase-3表达减少,抗凋亡蛋白Bcl-2表达增多。结论:Apelin-13可以有效缓解缺血引起的急性心肌梗死和心肌细胞凋亡,可以作为临床治疗急性心肌梗死的备选药物。
Objective:To study the protection effect of Apelin-13 on myocardial cell of acute myocardial infarction in rats.Methods:The acute myocardial infarction methods(AMI)model was developed by ligation of the left anterior descending coronary artery.The surviving rats were divided into AMI group(AMI)and Apelin-13 treated group(Apelin-13),and the groups of sham operation and Apelin-13 treated sham operation were also set up(n=8).After treatment for 14 d,the rat hearts were taken,then detected the myocardial infarction area by TTCEvans Blue staining and the pathological changes of cardiac tissue through HE staining.Furthermore,TUNEL method was used to detect the apoptotic myocardial cells,and the expression of the apoptotic protein Bcl-2,Bax and cleavedCaspase-3were detected by Western blotting.Results:Compared with the AMI group,after Apelin-13 treatment,the relative area of infarction myocardium in rats were significantly reduced,the accumulation of inflammatory cell in myocardial tissue and the number of apoptotic myocardial cells were decreased,the expression of pro-apoptotic protein Bax and cleaved-Caspase-3were increased and the expression of anti-apoptotic protein Bcl-2was decreased.Conclusion:Apelin-13 could effectively alleviate the acute myocardial infarction and myocardial cell apoptosis induced by myocardial ischemia,and Apelin-13 can be used as a drug for clinical treatment of acute myocardial infarction.
