康妇炎胶囊对兔盆腔炎性疾病后遗症模型粘连相关指标影响的实验研究 被引量:6

摘要 目的:通过观察康妇炎胶囊对盆腔炎性疾病(PID)后遗症模型兔子宫、输卵管组织粘连相关指标细胞间粘附分子-1(ICAM-1)、肿瘤坏死因子-α(TNF-a)、血管内皮生长因子(VEGF)表达的影响,从抗粘连方面探讨康妇炎胶囊治疗盆腔炎性疾病后遗症的机制。方法:将5~6个月龄新西兰雌兔分为:正常组、模型组、桂枝茯苓组、康妇炎组,除正常组外,其余3组均通过逆行宫腔插管灌注细菌法建立盆腔炎模型,造模成功后,模型组、桂枝茯苓组、康妇炎组分别灌服蒸馏水、桂枝茯苓胶囊、康妇炎胶囊3周,正常组灌服蒸馏水。肉眼观察各组兔子宫及输卵管情况;HE染色观察各组兔子宫、输卵管组织学变化;免疫组化及免疫印迹法检测各组兔子宫、输卵管组织ICAM-1、TNF-a、VEGF表达的变化。结果:肉眼观察模型组兔腹膜明显增厚,盆腔积液,子宫充血,质脆,弹性差,输卵管管壁粗糙、增厚,与周围组织粘连,或外观略呈串珠样;康妇炎组兔腹膜增厚不明显,无盆腔积液,子宫无明显充血,输卵管与周围组织无明显粘连;康妇炎组兔子宫、输卵管组织ICAM-1、TNF-a、VEGF表达较模型组明显降低(P〈0.05或P〈0.01)。结论:康妇炎胶囊治疗PID后遗症可能与其降调子宫、输卵管组织粘连相关指标ICAM-1、TNF-α、VEGF的表达有关。 Objective:To investigate the mechanism of Kangfuyan capsule treating sequelae pelvic inflammatory disease(PID)by observing the effect of Kangfuyan capsule on adhesion related indexs ICAM-l,TNF-α,VEGF in uterus and fallopian tube of rabbits with sequelae of PID.Method:New Zealand female rabbits with 5 ~ 6month age were divided into normal group,model group,Guizhifuling capsule group,Kangfuyan capsule group.Except the normal group,rabbits in other 3groups were all established the sequelae of PID model by injecting mixture of bacterial suspension into rabbits uterus.Model group,Guizhifuling capsule group,Kangfuyan capsule group were given distilled water,Guizhifuling capsule,Kangfuyan capsule for 3weeks respectively after the sequelae of PID models established successfully.The normal group also were given distilled water for 3weeks.We observed the pelvic,uterus,fallopian tubes situationg of all the rabbits with our naked eyes.HE staining to observe uterus,fallopian tubes pathological changes.Using immunohistochemistry method and western blot to detect the expression of the following indices in uterus and fallopian tubes of rats:ICAM-l,TNF-αand VEGF.Results:In the model group,peritoneal was thickened clearly,pelvic fluid was formed.Uterus were bloodshot,brittle and low elasticity.Fallopian tube walls were rough,thickened,had some adhesion with adjacent tissue,and showed string-of-beads pattern.In Kangfuyan capsule group,peritoneal was not markedly thickened,no pelvic fluid was found,uterus were no congestion,fallopian tubes were no adherence with surround-tissue.Compared with model group expression of ICAM-l,TNF-αand VEGF of Kangfuyan capsule group decreased significantly(P〈0.05 or P〈0.01).Conclusions:The mechanism of Kangfuyan capsule treating sequelae of PID might be related to that Kangfuyan capsule down regulating the expression of ICAM-l,TNF-αand VEGF in model rabbits.
