Ezrin和p16基因在宫颈癌和宫颈柱状上皮异位组织中的表达及其临床意义

Expression and clinical significance of Ezrin and p16 in tissues of cervical cancer and cervical columnar epithelium ectopy

目的探讨埃兹蛋白(Ezrin)和p16基因在宫颈癌和宫颈柱状上皮异位组织中的表达及其临床意义。方法采用实时逆转录-聚合酶链反应(RT-PCR)技术检测39例宫颈癌患者和35例宫颈柱状上皮异位患者组织Ezrin m RNA和p16 m RNA的表达,并选取正常宫颈组织作为对照组,比较分析以上指标的变化。结果 Ezrin m RNA和p16 m RNA在宫颈癌组、宫颈柱状上皮异位组及对照组中的表达率分别为89.7%、22.8%、21.0%和94.8%、22.8%、5.2%。Ezrin m RNA在宫颈癌组织中的表达率显著高于宫颈柱状上皮异位组及对照组(P<0.01),在宫颈柱状上皮异位与对照组中Ezrin m RNA表达率比较差异无统计学意义(P>0.05)。p16 m RNA在宫颈癌与宫颈柱状上皮异位组织中的表达率显著高于对照组(P<0.01),在宫颈癌组织的表达率高于宫颈柱状上皮异位组织,差异均有显著统计学意义(P<0.01)。Ezrin m RNA和p16 m RNA分别在宫颈癌组、宫颈柱状上皮异位组及对照组中的表达量分别为(0.82±0.17)、(0.25±0.06)、(0.23±0.07)和(0.77±0.12)、(0.35±0.07)、(0.22±0.05)。Ezrin m RNA和p16 m RNA在宫颈癌组中的表达量具有相关性(r=0.539,P<0.01)。结论检测组织Ezrin和p16基因表达可应用于宫颈癌和宫颈柱状上皮异位的鉴别诊断,并为宫颈癌的早期发现提供依据。

Objective To explore the expression and clinical significance of Ezrin and p16 in tissues for cer-vical cancer and cervical columnar epithelium ectopy. Methods The expression of Ezrin mRNA and p16 mRNA were detected with the polymerase reaction technology in the tissues of 39 patients with cervical cancer （group A） and 35 patients with cervical columnar epithelium ectopy （group B）. Normal cervical tissues were selected as the control group. The changes of above indicators were compared. Results The expression rates of Ezrin mRNA and p16 mRNA in group A, group B and the control group were 89.7%, 22.8%, 21.0%and 94.8%, 22.8%, 5.2%, respectively. The expression rates of Ezrin mRNA in group A was significantly higher than the other two groups （P〈0.01）, which showed no statistsically significantly differnece between group B and the control group （P〈0.05）. The expression rates of p16 mRNA in group A and group B were signifiantly higher than that in the control group （P〈0.01）, and that in group A was higher than that in group B （P〈0.01）. The expression quantities of Ezrin mRNA and p16 mRNA in group A, group B, the control group were （0.82±0.17）, （0.25±0.06）, （0.23±0.07） and （0.77±0.12）, （0.35±0.07）, （0.22±0.05）, respectively. Ex-pression of Ezrin mRNA and p16 mRNA were corelated in group A （r=0.539, P〈0.01）. Conclusion Testing the expres-sion of Ezrin and p16 in tissues can be used in the differential diagnosis between cervical cancer and cervical colum-nar epithelium ectopy, which provides basis for the early discovery of cervical cancer.
