外源物质对新疆紫草愈伤组织次生代谢物含量的影响 被引量:1

Effects of Exogenous Substances on the Content of Secondary Metabolites in Arnebia euchroma Johnst Callus

摘要 以新疆紫草愈伤组织为材料,研究外源物质AgNO_3、L-苯丙氨酸及乙酸钠对其次生代谢产物含量的影响。结果表明,M9培养基中添加AgNO_35 mg/L时,愈伤组织中多酚、花青素的含量分别较对照提高了2.0倍、2.9倍,紫草素及其衍生物、总黄酮的含量在AgNO_3浓度为3mg/L时达峰值,分别较对照提高了1.3倍、1.5倍。M9培养基中添加AgNO_35mg/L及L-苯丙氨酸25mg/L时,紫草素及其衍生物、多酚、花青素及总黄酮的含量均达峰值,分别较对照提高了3.4倍、3.0倍、10.9倍和2.3倍。在M9培养基中添加AgNO_35 mg/L、L-苯丙氨酸25mg/L基础上,乙酸钠处理的愈伤组织中的总黄酮、多酚、花青素、紫草素及其衍生物的含量均低于对照。在新疆紫草愈伤组织培养中添加诱导子和前体物质均可促进次生代谢产物的积累。 The effects of three factors (AgNO3 ,L-phenylalanine and sodium acetat)on the secondary me-tabolites content in callus of Arnebia euchroma Johnst were studied.The results showed that when 5 mg/L AgNO3 were added to M9 medium,the content of polyphenol and anthocyanin in callus respectively in-creased by 2.0,2.9 times compared with the CK.However,the content of shikonin and total flavonoid in callus respectively increased by 1.3,1.5 times when the AgNO3 concentration as 3 mg/L.When 5 mg/L AgNO3 were added to M9 medium and 25 mg/L L-phenylalanine,the content of shikonin,polyphenol,an-thocyanin and total flavonoid were the highest,respectively increasing by 3.4,3.0,10.9,2.3 times compared with the CK.When 5 mg/L AgNO3 and 25 mg/L L-phenylalanine were added to M9 medium,the content of total flavonoid,polyphenol,anthocyanin and shikonin in callus with different sodium acetate treatments were lower than the control.When the elictors and precursors were added to callus,secondary metabolites could be well accumulated.
