摘要
目的:完善甘草显微定量的方法。方法:在已有显微定量方法的基础上,以混悬制样代替"水飞"制样,以血球计数板代替普通载玻片;利用线性分析方法确定取样量,用正交试验方法,优化制片用混悬液配比和定量需要观察制片的数量,进行混悬液稳定性考察,确定实验用时。结果:样品细度要求100目,样品定量需制片10张,记录显微特征个数并计算。结论:引用血球计数板,提高显微定量的易操作性和准确性,方法是可行的。
Objective: To improve the micro-quantitative methods for licorice. Methods: Based on existing micro- quantitative methods, “ShuiFei” example preparation pattern (the process by which solid powder and liquid are grinded together in order to make suspension or remove contaminants ) replaced by suspending example preparation pattern, and substitute blood counting plates for microscope slides. Sampling number was determined by linear analysis. The proportion of suspension for plates making as well as the number of sampling plates needed to be observed to confirm the quantity were optimized by orthogonal experiment method, then the inspects on suspension stability was casted and experiments time was confirmed. Results: It turned out that sampling fineness should be 100 mesh, and determination of sample quantity needed 10 slides. After the preparation, the number of micro-characteristics of samples was counted and calculated. Conclusion: It is a practical way by using blood counting plates to increase the handle ability and accuracy of micro-quantitative method.
