葎草醇提物对小鼠细菌性腹泻的治疗作用

Therapeutic effect on Humulus scandens alcohol extract on bacterial diarrhea of mice

探讨葎草醇提物对金黄色葡萄球菌所致小鼠腹泻的治疗作用。金黄色葡萄球菌灌胃法制作小鼠腹泻模型,通过观察小鼠精神变化、体质量变化、DAI评分以及小肠病理切片变化的情况,评价葎草醇提物对小鼠的抗腹泻作用。高剂量治疗组与模型组相比,对小鼠精神恢复、体质量变化以及小肠绒毛恢复等均有明显的作用,高剂量组的治疗效果接近阳性对照组;高剂量治疗组和阳性对照组相对模型组体质量下降趋势明显降低,差异显著(P<0.05);与空白组相比,模型组DAI评分明显增加,差异显著(P<0.05)相对于模型组,高剂量组DAI、阳性治疗组DAI的评分明显降低,与空白组相比差异不显著(P>0.05),治疗效果显著,而中、低剂组DAI评分结果与空白组相比差异显著(P<0.05),治疗效果不明显。病理组织学观察,空白组小鼠小肠绒毛排列整齐,各层次结构清晰,绒毛间质无水肿、无小血管扩张充血,而模型组小鼠小肠绒毛排列紊乱,大部分绒毛脱落,绒毛表面有炎性渗出,绒毛间质水肿,小血管扩张、充盈。高剂量组和阳性对照组与模型组相比症状明显改变,小肠绒毛排列整齐,绒毛表面炎性渗出大部分消失,绒毛间质无明显水肿、无小血管扩张、充血,小肠结构与正常组织大致相同,治疗效果明显。葎草醇提物对小鼠腹泻有明显的治疗效果,可为细菌性腹泻的治疗提供理论依据。

To explore the therapeutic effect of Humulus scandens alcohol extract on diarrhea induced by Staphylococcus aureus in mice.The mental state,body weight,general morphological,histopathological changes were observed daily during the experiment and the disease activity index was detected to evaluate therapeutic action of Humulus scandens alcohol extract on diarrhea.Compared with normal group,the high dose group hada significantly change on metal state,body weight and small intestinal villus recovery,its treatment effect was close to the positive control group.Compared with model group,body weight decline of the high dose group and positive control group decreased markly and there was significant difference（P〈0.05）.Compared with normal group,the DIA score markly increased in model group,with significant（P〈0.05）the difference was.The high dose group and positive control group decreased DAI compared with model gropup,which were no significant difference（P〉0.05）with the normal group,the treatment effect was remarkable;while the mid and low does group were significant difference with normal group（P〈0.05）,the treatment effect was not ideal.Histological examination showed that the normal group of mice with small intestinal villus well-arranged,the hierarchical structure is clear,and there was no small vessel hyperemia and no villous intestinal edema.While small intestinal villus of model group disordered,most of the villus fell off,there was inflammation exudation on the surface of villi and villous edema and congestion of small blood vessels.Compared with the model group,highdose group and positive control group greatly reduced cell inflammatory exudation,edema and congestion.Humulus scandens alcohol extract hada significant therapeutic effect on diarrhea in mice,which provide a theoretical basis for the treatment of bacterial diarrhea.
