摘要 目的:用星点与正交设计两种方法优化穿龙薯蓣总皂苷的提取工艺。方法:采用回流法提取穿龙薯蓣总皂苷,以薯蓣皂苷为对照品,以乙醇浓度、提取时间、提取温度和料液比为考察指标,通过UV法测定其总皂苷含量。结果:最终确定穿龙薯蓣总皂苷的最佳提取工艺为:乙醇浓度为75%,料液比为16,提取时间为130 min/次,提取温度为85℃。结论:该方法设计精确,数据可靠,可为穿龙薯蓣总皂苷的提取提供了理论依据。 Objective:To optimize the process of total sapogenin extracted from dioscorea nipponica makino with central composite design and orthogonal experimental design. Methods:The refluxing extraction method of total sapogenin was adopted,dioscin as reference substance,with ethanol concentration,extraction time,extraction temperature and solid-liquid ratio as examining indice. The content of sapogenin was measured by UV method. Results:The optimum extraction conditions of sapogenin were as follows,75% ethanol concentration,solid-liquid ratio was 16,the extraction time was 130 min per time,extraction temperature was 85 ℃. Conclusion:This experimental design method is precise and the data is reliable. It can provide a theoretical basis for the extraction of total sapogenin extracted from dioscorea nipponica makino.
