摘要
目的:建立离子对反相高效液相色谱法测定血浆中百草枯的浓度。方法:采用Discovery C_(18)色谱柱;流动相为乙腈-庚烷磺酸钠溶液(12∶88);10%三氯醋酸直接沉淀血浆蛋白;流速为1.0 m L·min^(-1);检测波长为258 nm。结果:定量下限为0.1μg·m L^(-1),线性范围为0.1~10.0μg·m L^(-1),批内和批间RSD分别为3.77%和10.60%,平均回收率大于89.31%。结论:本法具有灵敏、准确、稳定、操作简便等优点,符合生物样本测定的要求,适用于测定人血浆中百草枯的浓度。
Objective: To establish a method for determination of paraquat in plasma by reversed phase ion-pair HPLC. Methods: The Discovery C_(18) column was used for separation and the mobile phase consisted of a mixture of acetonitril-sodium heptanesulfonate solution(12 ∶88, v ∶v). The plasma proteins were precipitated by 10% trichloroacetic acid. The flow-rate was 1.0 m L ·min^(-1), and the detection wavelength was 258 nm. Results: The detection limit of the method was 0.1 μg·m L^(-1). The linear calibration curves were obtained in concentration range of 0.1-10.0 μg·m L^(-1). The intra- and inter-day RSD were3.77% and 10.60%, respectively. The average recovery was above 89.31%. Conclusion: The method is simple and rapid, which can be used for determination of paraquat in human plasma.
