摘要
目的探讨外伤性脑损伤患者伤后血清及脑脊液中Nogo-A基因表达和脑损伤严重程度的关系。方法根据格拉斯哥昏迷评分,60例急性闭合性颅脑损伤患者均分为轻、中、重三组,于入院第1、4、7和10天分别采集静脉血和脑脊液,检测Nogo-A mRNA和蛋白的表达。结果重度组脑脊液中Nogo-A mRNA的含量在各时间点均高于轻度组(P<0.01);血清中Nogo-A mRNA表达量在第4、7和10天高于轻度组(P<0.05、P<0.01和P<0.05)。重度组患者脑脊液中Nogo-A蛋白的表达量在第4、7和10天高于轻度组(P<0.01);血清中Nogo-A蛋白含量在第7和10天高于轻度组(P<0.01)。结论相对于外周血而言,脑脊液中Nogo-A mRNA的水平变化与临床病情相一致。
Objective To investigate the relationship between the expressions of Nogo-A gene in CSF and serum and the severity of traumatic brain injury.Methods According to Glasgow coma scores,60 patients with traumatic brain injury were equally assigned into 3 groups of A(mild),B(moderate)and C(severe).CSF and serum samples were collected on the 1st,4th,7th and 10 th day after trauma for detecting the mRNA and protein expressions of Nogo-A.Results The expression of Nogo-A mRNA in CSF after trauma was significantly higher in group C than that in group A at all time points(P〈0.01),which in serum was significantly higher in group C than that in group A on the 4 th,7th and 10 th day after trauma(P〈0.05,P〈0.01 and P〈0.05).The expression of Nogo-A protein in serum after trauma was significantly higher in group C than that in group A on the 4th,7th and 10 th day after trauma(P〈0.01),which in serum was significantly higher in group C than that in group A on the 7th and 10 th day after trauma(P〈0.01).Conclusion Compared to the expression of Nogo-A mRNA in serum,the changes of Nogo-A mRNA expression in CSF are more consistent with the severity of traumatic brain injury.
