初步探求酶标记抗体的合适比例

摘要 目的探求酶标记抗体时的最佳比例,以用于体外诊断的实践。方法辣根酶与CEA抗体按照质量比10∶1~1∶10进行连接,着重考察各酶结合物的样品检测情况和在洗涤过程中游离酶的干扰规律,最终找出了酶标记抗体的适宜趋势。结果当抗体与辣根酶的质量比达到5到10倍时,计数高于S0的样品的个数最多,实现了通过标准曲线方程反算出浓度的理想状态。结论在标记抗体的过程中,只需五分之一甚至十分之一的酶量已能够满足需求,过多反而会增加干扰的风险。 Objective To explore the best labeled proportion of enzyme and antibody for in vitro immunoassay.Methods The CEA antibody was labeled with Horseradish peroxidase at the mass ratio of 1 ∶ 10 to 10 ∶ 1. The various conditions were studied to optimize the appropriate proportion of enzyme and antibody tendency for the labeling. Results When the antibody to Horseradish peroxidase weight ratio reached 5-10 times,there were more samples whose count higher than that of S0 could be detected. It was the ideal state to calculate samples concentration based on standard curve equation. Conclusion In the antibody labeling process,one fifth or even one tenth of enzyme amount could be sufficient for the labeling to avoid the risk of interference for in vitro immunoassay.
