摘要
从质粒pJSB15中分离出人IL-2RαcDNA 673b_p片段,用放射性同位素α-^(32)p标记此片段,制备了IL-2RαcDNA探针。利用该探针与淋巴细胞RNA进行点杂交。结果证明,pHA、TPA均可诱导IL-2Rα转录增加,二者合用有明显的协同效应;且发现新鲜分离的多种白血病细胞测出的IL-2R_α(?)RNA水平高低不同,这可能有助于临床分型。
We obtained human IL-2R cDNA 673 bp fragments from plasmid pJSB15 and prepared α-32 p-IL-2 R cDNA probe. Dot hybridizations of RNA extracted from human lymphocytes were done by using the probe. The results showed that PHA or TPA could induce IL-2 R mRNA, respectively; and the synergistic effects were seen when used them together. IL-2R mRNA could be expressed spontaneously in many kinds of leukemic lymphocytes isolated freshly from PBL of leukemic patients.
