七氟醚后处理对成熟兔心肌缺血再灌注后IL-8、IL-10浓度及心肌组织超微结构的影响 被引量:1

Effect of Sevoflurane Postconditioning on the Levels of IL-IL-8 and IL-IL-10 and on the Ultramicrostructure of Myocardial Tissue after Myocardial Ischemia Reperfusion in Mature Rabbits

摘要 目的研究七氟醚后处理对成熟兔心肌缺血再灌注后IL-8、IL-10浓度及心肌超微结构的影响。方法选32只新西兰成熟兔随机分为缺血再灌注组(I/R组)与七氟醚后处理组(S组),每组16只。I/R组结扎冠状动脉前降支缺血30 min,再灌注240 min。S组结扎冠状动脉前降支缺血30 min,再灌注即刻吸入2.5%七氟醚30 min,洗脱10 min,再灌注持续至240 min。监测缺血前(T1)、再灌注后1h(T2)、2h(T3)、4h(T4)血浆IL-8、IL-10浓度,观察心肌超微结构变化。结果 T2~T4时两组IL-8、IL-10均高于T1时(P〈0.05),两组组内T1时分别比较IL-8、IL-10,差异无统计学意义(P〉0.05);S组T2~T4时IL-8表达明显低于IR组(P〈0.05),而IL-10表达明显高于IR组。显微镜下S组超微结构损伤较I/R组轻。结论七氟醚后处理可抑制缺血再灌注后心肌致炎性介质IL-8的过渡表达,增强抗炎性介质IL-10的表达,调节致炎抗炎平衡,减轻缺血再灌注心肌超微结构的损伤。 Objective To explore the effect of sevoflurane postconditioning on the levels of IL-8 and IL-10 and on the ultramicrostructure of myocardial tissue after myocardial ischemia reperfusion(MIR) in mature rabbits. Methods 32 mature New Zealand rabbits were randomly divided into 2 groups: I/R group(ischemia reperfusion group) and S group(sevoflurane postconditioning group), 16 in each; the anterior descending coronary artery occlusion for 30 minutes followed by reperfusion for 240 minutes was made to rabbits in I/R group while to rabbits in S group, the anterior descending coronary artery occlusion for 30 minutes followed by reperfusion for 30 minutes with immediate inhalation of 2.5% sevoflurane was first made and then an elution for 10 minutes followed by reperfusion for240 minutes; a detection was made to the level of IL-8 and IL-10 at the time point of before ischemia(T1), 1 hour after reperfusion(T2),2 hours after reperfusion(T3) and 4 hours after reperfusion(T4) while an observation was made to the changes of ultramicrostructure of myocardial tissue at each time point in both groups. Results The levels of IL-8 and IL-10 at T2 and T4in both groups were higher than those at T1(P〈0.05) while no difference of statistical significance was found in the levels of IL-8 and IL-10 at T1 from those at T2 and T4in each group; the levels of IL-8 at T2 and T4in S group were much lower than those in I/R group((P〈0.05) while the level of IL-10 in S group was much higher than that in I/R group; microscopic examination showed a slighter myocardial ultramicrostructural injury in S group than in I/R group. Conclusions Sevoflurane postconditioning can inhibit the over-expression of IL-8, enhance the expression of IL-10, regulate the balance of inflammatory and anti-inflammatory cytokines expression and reduce the myocardial ultramicrostructural injury after ischemia-reperfusion.
