摘要
详细描述了利用蛋白酶解法和葡聚糖酶解法检测酵母β-葡聚糖的方法。并对该方法进行了优化,最后验证了测定方法的精密度,回收率及日间重复性实验。实验结果显示,该检测方法测得的酵母β-葡聚糖质量分数的精密度RSD为1.5%,平均回收率均在87%~98%之间,日间重复性稳定。此方法适用于作为牛奶中酵母β-葡聚糖特别是[(1-3)(1-6)-β-glucan)]质量分数的测定方法。
The content of yeast β-glucan added in milk was quantified by using protease hydrolysis with glucanase hydrolysis method. The :ondition of this method was also improved. The precision and recovery of the test was investigated. The results showed that the RSD value 3fprecision test was 1.5%. The average recovery ranged from 87% to 98%, and the intra-day and inter-day assay was stable. In conclusion, the developed method allows yeast β-glucan quantification (specifically for mixed-linkage (1-3)(1-6)-β-glucan)to be conducted in milk.
