摘要
目的研究大蒜素干预下SD大鼠血清诱导食管癌EC109细胞凋亡及其对Survivin蛋白表达的影响。方法Ⅰ组:以普通灭活SD大鼠血清培养液培养食管癌EC109细胞;Ⅱ组:以大蒜素干预SD大鼠灭活血清培养液培养食管癌EC109细胞;采用AnnexinⅤ-EGFP/PI双染法和亚倍体峰法应用流式细胞仪检测2组食管癌细胞早期和中晚期凋亡率;Survivin蛋白的表达应用Western blot方法检测。结果Ⅱ组细胞凋亡率明显高于Ⅰ组,差异有统计学意义(P<<0.05);Ⅱ组细胞中Survivin蛋白的表达水平明显低于Ⅰ组,差异有统计学意义(P<0.05)。结论长期大蒜素干预下SD大鼠血清能促进食管癌EC109细胞的凋亡并抑制Survivin蛋白的表达。
Objective To investigate the effect of SD rats serum after intervention by allicin on the apoptosis of esophagus cancer EC-109 cells cell and the expression levels of Survivin. Methods The group Ⅰ: esophagus cancer EC109 cells were cultured in normal inactivated SD rat serum medium; group Ⅱ: esophagus cancer EC109 cells were cultured in inactivated SD rat serum medium plus allicin,then the cell apoptosis rates were detected by Annexin Ⅴ-EGFP / PI double-standing and hypoploid peak method with flow cytometer,furthermore the expression levels of Survivin were measured by Western Blot. Results The cell apoptosis rates in group Ⅱ were significantly higher than those in group Ⅰ group( P〈 0. 05),however,the expression levels of Survivin protein in group Ⅱ were significantly lower than those in group Ⅰ( P 〈0. 05). Conclusion The serum of SD rat after long-term intervention by allicin can promote the apoptosis of esophagus cancer EC109 cells and can inhibit the expression of Survivin protein.
