摘要
目的探讨表阿霉素对不同乳腺癌细胞系的作用效应。方法应用流式细胞仪检测并分析表阿霉素作用于乳腺癌细胞系MCF-7、SKBR3、MDA-MB-231及MDA-MB-468后免疫原性标志物的表达以及不同乳腺癌细胞系死亡过程中免疫原性死亡的能力;采用CCK-8方法观察表阿霉素对不同乳腺癌细胞系增殖的抑制效应。结果 (1)表阿霉素作用于各个乳腺癌细胞系24 h后,MDA-MB-231与MDA-MB-468细胞系的增殖率明显低于MCF-7细胞系的增殖率,差异有统计学意义(P<0.05)。(2)MDA-MB-231与MDA-MB-468细胞系表阿霉素作用24 h的IC50值明显低于MCF-7与SKBR3细胞系。(3)钙网蛋白、HSP70随着表阿霉素作用时间的增加而增加,24h后MDA-MB-231与MDA-MB-468细胞系中的表达明显高于MCF-7细胞系,差异有统计学意义(P<0.001)。SKBR3细胞系中钙网蛋白、HSP70的表达则稍高于MCF-7细胞系(P>0.05)。结论表阿霉素的作用效应不仅包括细胞毒性作用,还潜在免疫原性死亡作用过程;激素受体阴性乳腺癌细胞死亡过程中的免疫原性死亡作用较强。
Objective The effect of Epirubicin on different breast cancer cell lines. Methods The human breast cancer cell lines MCF7,SKBR3,MDA-MB-231 and MDA-MB-468 were treated with Epirubicin respectively,and the markers of immunogenic cell death were detected by flow cytometry; the proliferation of cell was tested by CCK-8 method. Results( 1) After treating for 24 h,the proliferation of MDA-MB-231 and MDA-MB-468 was lower than MCF-7( P〈0. 005).( 2) IC50 of Epirubicin to MDA-MB-231 and MDA-MB-468 was lower than MCF-7 and SKBR3.( 3) The amount of calreticulin and HSP70 was increased gradually with the treatment of Epirubicin; after24 hours,these markers of MDA-MB-231 and MDA-MB-468 were significantly higher than that of MCF-7( P〈0.001). The both markers of SKBR3 were higher than that of MCF-7,but not significantly( P〉0. 005). Conclusion The effect of Epirubicin not only includes cytotoxic effect,but also potential immunogenicity death process. During the process of cell death,the immunogenicity death effect of hormone receptor negative breast cancer cell is stronger.
