摘要 为了优选烟草(Nicotiana tabacum L.)提取物最佳提取方法,采用高效液相色谱法测定了不同条件下超声波制备的烟草提取物。结果表明,在色谱柱为Inertsil ODS-SP(4.6 mm×250 mm,5μm),流动相为甲醇-磷酸缓冲液(1∶9),检测波长为259 nm的条件下,通过测定和比较烟草在不同溶剂(水、甲醇)、不同提取时间、不同Na OH浓度、不同超声波频率、不同品种条件下的特征色谱峰情况及参照峰烟碱含量,筛选出以0.5%Na OH水溶液,40 k Hz超声波提取30 min为烟草提取物最佳制备工艺,且选择红花烟叶较好,为制备烟草提取物和烟草烟油替代品奠定了基础。 In order to select the best method for extracting tobacco, the method of HPLC was applied to assay the HPLCs of tobacco extracts from different extraction conditions in ultrasonic process. The results showed that : Under the condition that the separation was performed on a Inertsil ODS-SP column(4.6 mm ×250 mm, 5 μm) with a mobile phase consisting of methanol and phosphate buffer solution(1∶9) and the detection was carried out on a UV detector at 259 nm, the characteristic fingerprints and nicotine content of reference peaks in the conditions of different solvent(water or methol), time of extraction, Na OH concentration, ultrasonic frequency and source(Tongliao or Lianyungang) were assayed and compared. The selected process was tobacco extracted with ultrasonic wave at 40 k Hz for 30 min in water with 0.5% Na OH, and the tobacco from Nicotiana tabacum L. was better, which provides basis for the preparation tobacco extraction and tobacco oil alternative.
