摘要
目的:探讨糖痹康对糖尿病周围神经病(diabeticperipheralneuropathy,DPN)小鼠感觉神经功能的影响。方法:将60只8~9周龄的ob/ob小鼠随机分为5组,另选取12只同龄C57BL/6J小鼠为对照组,分别给予甲钴胺、糖痹康及溶媒8周。监测体质量、50%机械缩足阈值(mechanicalwith—drawthreshold,MwT)、甩尾反射潜伏期(tail-flicklatency,TFL)、感觉神经传导速度(sensorynerveconductionvelocity,SCV)。结果:与对照组比较,给药后模型组体质量显著增加(P〈0.001),50%MWT显著下降(P〈O.05),TFL显著延长(P〈0.05),SCV显著减慢(P〈0.05);与模型组相比,各给药组体质量未见显著差异;给药4周时,糖痹康高剂量组50%MWT明显上升(P〈0.05),糖痹康高、低剂量组及甲钴胺组TFL显著缩短(P〈0.05);给药8周时,糖痹康高、中剂量组及甲钴胺组50%MWT明显上升(P〈0.05),糖痹康高、低剂量组及甲钴胺组TFL显著缩短(P〈0.05),糖痹康中剂量组及甲钴胺组SCV显著上升(P〈0.05)。结论:ob/ob品系小鼠模型适合于药物对DPN疗效的评价,糖痹康能改善DPN小鼠的感觉神经功能。
Objective:To investigate the influence of Tang Bi Kar.g on sensory neuropathy of dia- betic peripheral neuropathy(TPN) mice. Methods:Sixty ob/ob mice of 8--9 week-old were randomly di- vided into five groups,other 12 age-matched C57BL/6J mice were defined as the control group, which were administered with mecobalamin, Tang Bi Kang and dissolvent. Body weight, 50% mechanical with- draw threshold(MWT),tail-flick latency(TFL),sensory nerve conduction velocity(SCV) were measured on time. Results:Compared with the control group, mice in the model group showed significant increase in weight (P〈0. 001), significantly decrease in 50% MWT(P〈0.05), significant extension in TFL(P〈 0.05) ,and obvious declineing in SCV(P〈0.05). Compared with the model group, the changes of body weight in each group were not obvious. After administration of drugs for 4 weeks,50% MWT increased significantly in the high dose group(6 g/kg Tang Bi Kang), the TFL was shortened significantly in the high dose group(6 g/kg Tang Bi Kang) and in the low dose group(1.5 g/kg Tang Bi Kang)and in the mecobalamin group. After administration of drugs for 8 weeks, 50% MWT increased significantly in the high dose group, the middle dose group(3 g/kg Tang Bi Kang) and the mecobalamin group. TFL was shortened significantly in the high dose group , the low dose group and the mecobalamin group,and SCVincreased significantly(P〈0.05) in the middle mice can be used to evaluate the medicinal cure / ropathy of diabetic peripheral neuropathy mice. dose group and the mecobalamin group. Conclusion:The effect for DPN, Tang Bi Kang may improve sensory neuropathy of diabetic peripheral neuropathy mice.
