Effect of blended canola oil and lard on the PPARα gene expression in liver of mice

摘要 为了探讨日粮中加入菜籽油和猪油调和油对小鼠肝脏PPARα基因表达的影响,试验将雄性KM小鼠随机分成正常剂量组、高剂量组,分别在日粮中添加3.8%和6.5%调和油,8周后称量体重,取完整肝脏组织,计算肝指数,H.E.染色进行病理观察,实时荧光定量RT-PCR检测肝脏PPARα水平。结果表明:与正常剂量组比较,高剂量组小鼠肝脏病理剖检无眼观病理变化,体重、肝功能无统计学意义(P>0.05),肝脏PPARα水平极显著上升(P<0.01)。说明饮食中过量摄入菜籽油和猪油调和油极显著上调小鼠肝脏PPARα的表达。 To investigate the effect of the addition of blended eanola oil and lard to the diet on the peroxisome proliferator - activated receptor α (PPARα) gene expression in livers of mice, the male KM mice were randomly divided into normal dose group and high dose group. The 3.8% and 6.5% blended oil was added in the diet, respectively, The body weight of mice was weighed after eight weeks, and the complete liver tissues were taken for calculating liver indexes. Hematoxylin and eosin ( H. E. ) staining was used for pathological observation, and the PPARα expression level in liver was detected by real - time fluorescence quantitative RT - PCR. The results showed that compared with the normal group, no pathological change was visually found in the pathological necropsy examination of livers in the mice from the high dose group, and no statistical significance was found ( P 〉 0.05 ) in body weight and river function indexes. However, the PPARα mRNA expression level in liver was significantly (P 〈 0.01 ) increased in the high dose group. The results indicate that excessive intake of dietary blended eanola oil and lard can significantly upregulate the expression of PPARα in livers of mice.
