摘要
本实验旨在探讨0~28日龄樱桃谷鸭的羽色变化及换羽的分子机制,用荧光定量PCR方法研究了酪氨酸酶基因(TYR)与血管内皮生长因子A基因(VEGFA)的m RNA表达谱。结果表明:0~7日龄绒羽色由黄变浅,21~28日龄绒羽逐渐变白并被幼羽取代;随着日龄增长,TYR基因在心、肝的表达量被抑制(P〈0.05),而眼、肌肉、脑则大量表达(P〈0.01);皮肤TYR基因呈先升后降的趋势,在肺、肾的表达则是先升后降再升的波动表达。VEGFA基因在0~7日龄樱桃谷鸭的肝、皮肤、肺、肾、眼、肌肉中表达明显增加(P〈0.01),而心、脑降低(P〈0.05);21~28日龄,在心、肝、皮肤、肺、肾VEGFA表达下调(P〈0.01),在眼、肌肉、脑中的表达却上调(P〈0.05)。综上,0~28日龄的樱桃谷鸭羽毛颜色和换羽与TYR基因和VEGFA的抑制表达有关,两者之间有一定协同性。
In order to study the molecular mechanism of duck feather color and molting development, the mRNA expression profiles of tyrosinase gene (TYR) and vascular endothelial growth factor A (VEGFA) gene were studied by quantitative real-time PCR in the cherry valley duck from 0 to 28 days old. The results showed that deep yellow down feathers changed shallow yellow feathers from 0 to 7 days, and down feathers gradually became white and were replaced by chick from 21 to 28 days old. Along with days of age growth , the expression of TYR gene was inhibited in heart and liver (P〈0.05), while up-regulations were observed in eyes, muscles and brain (P〈0.01). Up-down expression profiles was detected in skin, and up-down-up expression profiles were occurred in the lung, kidney. During 0-7 days old, the expression of VEGFA gene extremely increased in liver, skin, lung, kidney, eye and muscle (P〈0.01), but decreased in heart and brain (P〈0.05). From 21 to 28 days old, the expression of VEGFA gene reduced in heart, liver, skin, lung and kidney (P〈0.01), while up-regulated in eye, muscle and brain (P〈0.05), respectively. Overall, the decreased expression profiles of TYR gene and VEGFA gene showed some convergence associated with the feather color and molting in the Cherry Valley duck during 21 to 28 days old.
