裸鼠肺癌原位模型的建立及螺旋CT、近红外荧光成像评价 被引量:3

Establishment of orthotopic lung cancer model in nude mice and its evaluation by spiral CT and near-infrared fluorescence imaging

摘要 目的 利用A549细胞系建立原位肺癌的动物模型,并采用螺旋CT扫描和近红外荧光成像评价胸内肿物生长情况。方法 将A549细胞悬液(5×106 ml-1)接种于20只裸鼠胸腔内,分别于接种后第7天、14天、21天,连续3周对荷瘤裸鼠行螺旋CT检查,动态观察胸内肿瘤生长情况,主要观察裸鼠的生存时间、体重变化及胸腔内肿瘤浸润和转移情况,并对裸鼠进行解剖后行近红外荧光成像和病理切片检查。结果 本次A549细胞的造模成功率为90%,中位生存期为30.7(20~41)天。荷瘤裸鼠胸内肿瘤转移发生率100%。经螺旋CT扫描肿瘤显示率100%,CT上测得平均肿瘤直径随时间逐步递增,而近红外荧光成像对胸腔内肿瘤显示率为零。结论 A549细胞接种于裸鼠肺部所建立的肺癌原位模型简单,成功率高;采用螺旋CT扫描能动态直观评价裸鼠胸内肿物生长情况,近红外荧光成像对胸腔内肿瘤显示能力有限。 Objective To establish orthotopic lung cancer animal model of the A549 cell line and to evaluate the growth pattern of orthotopic lung cancer tumors by spiral CT and near-infrared fluorescence imaging.Methods A549cells(5×106 ml-1)were suspended and inoculated into the left lung of BALB/c nude mice(20cases)by the intrathoracic injection method.Nude mice were scanned three times by spiral CT after inoculated A549 cell line every week.The survival-time,weight,tumor invasion and metastasis of the mice were examined.The tissue was collected for the subsequent near-infrared fluorescence imaging and histology assay after autopsia the mice.Results Tumor-form-rate of the orthotopic lung cancer animal model was 90%.The median survival time was 30.7(20~41)days.The tumor metastasis incidence was100%.All tumors appeared on spiral CT.The mean tumor diameter gradually increased with time.Tumors had not been found by near-infrared fluorescence imaging.Conclusion The lung cancer orthotopic model,which is established by A549 cells transplanted into the lung of the nude mice,is simple and high successful.The spiral CT can be used to evaluate thoracic tumor growth in nude mice visually and dynamically,the near-infrared fluorescence imaging can not do it.
