摘要
目的探讨丹红注射液对大鼠蛛网膜下腔出血(SAH)后脑血管痉挛(CVS)的防治作用。方法将84只健康成年SD大鼠随机分成4组:正常组(n=12)、生理盐水组(n=24)、SAH组(n=24)和丹红组(n=24);后3组根据造模后取材时间,各分为造模后3 d和7 d两亚组,每亚组12只大鼠。采用枕大池二次注血法建立大鼠SAH模型。丹红组在造模后每天给予一次丹红注射液腹腔注射治疗。按上述时间点灌注固定后取大鼠脑桥段基底动脉,行HE染色,400倍光镜下观察,并测量基底动脉内径、血管壁厚度。结果造模后3 d、7 d,生理盐水组基底动脉内径、管壁厚度与正常组均无明显变化(P>0.05)。造模后3 d,与生理盐水组相比,SAH组和丹红组基底动脉内径明显缩小,管壁厚度明显增加(P<0.05);但SAH组和丹红组之间无统计学差异(P>0.05)。造模7 d后,SAH组基底动脉内径近一步缩小(P<0.05),管壁厚度进一步增加(P<0.05);而丹红组基底动脉内径较SAH组明显增加(P<0.05),管壁厚度明显变薄(P<0.05)。结论丹红注射液对大鼠SAH后CVS有一定的缓解作用。
Objective To explore the preventive and therapeutic effects of Danhong on the cerebral vasospasm (CVS) after subarachnoid hemorrhage(SAH) in rats. Methods Eighty-four healthy SD rats were divided into 4 groups, i.e. the normal control group (n=12), saline group (n=24, where saline was injected into the cisterna magna), SAH group (n=24) and Danhong group (n=24, where the rats with SAH weretreated by Danhong). Saline, SAH and Danhong groups were divided again into two subgroups respectively, i.e. the saline subgroups 3 and 7 days after the second injection of the blood. These subgroups included 3 and 7 days subgroups, 3 and 7days SAH sub-groups, and 3 and 7 days Danhong subgroups. There were 12 rats in each subgroup. The model of SAH was established by two injections of autogenic blood into the cerebellar cisterna magna. The changes in the morphology of the basilar arteries were observed and the inner diameter and vascular walls thickness of the basilar arteries were determined in all the groups. Results The inner diameter and the vascular walls thickness of the basilar arteries were (210.01 ±20.62)μm and (15.2±0.58)μm respectively in the 3 days SAH group, and their inner diameter and vascular wails thickness were (218.07±21.85)μm and (14.32±0.37)μm respectively in 3 days Danhong group. The inner diameter and vascular walls thickness of the basilar arteries were (157.71±25.13) μm and (18.3±0.45)μm respectively in7 days SAH group, and their inner diameter and vascular walls thickness were (222.38±17.29)μm and (12.67±0.55)μm respectively in 7 days Danhong group. There were significant differences in the inner diameter and vascular walls thickness of the basilar arteries between 3 days SAH and Danhong sub-groups, and 7 days SAH and Danhong sub-groups. Condusion it is suggested that Danhong has a role in the mitiKation of CVS after SAH in rats.
