摘要
【目的】了解和研究广西地方柑橘资源的亲缘关系和遗传多样性。【方法】以10份外省野生和地方柑橘种质为参照,采用ISSR分子标记技术对10份广西地方柑橘种质和8份姑婆山野生柑橘资源的遗传多样性及其亲缘关系进行分析和评价。【结果】从100条ISSR引物中筛选出12条引物对供试材料进行多态性分析。获得总条带67条,其中多态性条带62条,多态位点百分率为92.54%。聚类结果显示28份资源可分为7个组,其中广西资源分布于6个组,姑婆山野生柑橘被聚为同一组,进而可划分为2个亚组。【结论】ISSR标记能较好地区分广西地方柑橘资源,且广西柑橘地方资源遗传多样性丰富。
[Objective] To elucidate the genetic relationship and genetic diversity of Guangxi local Citrus resources. [Methods] In this study, ISSR molecular marker techniques was used to detect the genetic diversity and relationship of 10 local Citrus and 18 wild Citrus and resources were using. [Results]A total of 67 bands were generated from 12 ISSR primers, of which 92.54% was polymorphic. As analyzed by NTSYS-pc, all the 28 resources can be divided into seven groups, and parts of Guangxi resources into six groups. Eight Guposhan wild Citruses are clustered into the same group, and thus can be divided into two subgroups. [Conclusion] Guangxi local Citrus can be distinguished by ISSR markers, and the genetic diversity of citrus in Guangxi is rich.
