滑膜炎颗粒对大耳白兔膝关节损伤保护作用的实验研究 被引量:3

Protective effect of Huamoyankeli on the knee osteoarthritis model of rabbit

摘要 目的观察滑膜炎颗粒对大耳白兔膝骨关节炎(kneeosteoarthritis,KOA)模型的疗效,并探讨其可能的作用机制。方法将大耳白兔按随机数字表法分为正常对照组,假手术组,模型组,壮骨关节丸组,美洛昔康组及滑膜炎颗粒大、中、小剂量组,除对照组及假手术组外,其他各组均采用改良Hulth法建立兔KOA模型,取兔左膝关节内侧入路,切断内侧副韧带,摘除内侧半月板,切断前交叉韧带,假手术组只暴露然后缝合。壮骨关节丸组灌胃壮骨关节丸混悬液0.56g/kg,美洛昔康片组灌胃美洛昔康片混悬液O.70mg&g,滑膜炎颗粒大、中、小剂量组分别灌胃滑膜炎颗粒混悬液5.02、2.50、1.26g/kg。正常对照组、假手术组及模型组灌胃等体积生理盐水。1次,d,连续给药4周。每周测体质量并进行关节肿胀评分,4周后,取各组动物的膝关节组织进行组织病理切片,光镜下观察。结果给药后2、3、4周,与模型组比较,滑膜炎颗粒小、中剂量组肿胀评分[(1。33±0.49)分、(1.42±0.51)分比(1.83±0.39)分;(1.42±0.51)分、(1.42±0.51)分比(1.92±0.51)分;(1.83±0.58)分、(1.42±0.51)分比(2.33±0.65)分]明显降低(P〈0.01或P〈0.05)。与模型组比较,滑膜炎颗粒大剂量组大耳白兔膝关节上皮增生[(0.91±0.54)分比(1.83±0.72)分]改善,滑膜内层炎细胞浸润[(0.09±0.30)分比(0.08±0.29)分]减少,滑膜下层纤维增生[(1.45±0.69)分比(2.00±1.04)分]、血管化[(1.45±0.52)分比(2.00±1.04)分]减轻,总评分[(11.00±2.72)分比(15.17±6.51)分]降低(P〈0.01或P〈0.05)。结论滑膜炎颗粒对KOA模型大耳白兔膝关节损伤有较好的保护作用。 Objective To observe the effect and mechanism of Huamoyankeli on the knee osteoarthritis model of rabbit. Methods Osteoarthritis models were established by improved Hulth method. Rabbits were randomly divided into 8 groups:the normal group, sham operation group, model group, zhuangguguanjiewan group, meiluoxicam group, Huamoyankeli high dose group, Huamoyankeli middle dose group and Huamoyankeli low dose group. Measuring the weight and grade the swelling of knee-joint every week. Four weeks later, we take out the knee-joint of all animals in each group, make slides and histopathological observation under light microscope. Results After taken drugs for more than two weeks, the animals in treated groups improved the score of swelling. Compared with the model group, the scores of thr Huamoyankeli low dose group and the middle dose group (1.33 ± 0.49, 1.42 ±0.51 vs. 1.83 ± 0.39; 1.42 ± 0.51, 1.42 ± 0.51 vs. 1.92 ± 0.51; 1.83 ± 0.58, 1.42 ± 0.51 vs. 2.33 ±0.65) were decreased at the second, third and fourth week(P〈0.01 or P〈0.05). The average weight of animals with operation was higher than the normal group. Observed under light microscope, slides of the Huamoyankeli group had different degrees of improvement, the epithelial hyperplasia (0.91± 0.54 vs. 1.83 ±0.72) improved, the inflammatory cells infiltration (0.09 ± 0.30 vs. 0.08 ±0.29), the fiber hyperplasia(1.45 ±0.69 vs. 2.00 ± 1.04), the vascularization (1.45± 0.52 vs. 2.00 ± 1.04) and the total score (11.00 ± 2.72 vs. 15.17± 6.51) reduced (P〈O.O1 or P〈O.05). Conclusion Huamoyankeli had protective effect on rabbit model of osteoarthritis.
