姜黄素缓解2型糖尿病小鼠肝脏的代谢紊乱 被引量:3

摘要 目的:研究姜黄素(curcumin)对2型糖尿病(type 2 diabetes mellitus,T2DM)小鼠肝脏代谢紊乱的影响。方法:建立T2DM小鼠模型,将造模成功的T2DM小鼠随机分成姜黄素给药组(50 mg/kg、100 mg/kg)和模型组,正常小鼠作为阴性对照组。通过生化指标检测、酶联免疫吸附、免疫印迹、和苏木素-伊红染色实验,探讨姜黄素对糖尿病肝脏的药理活性。结果:同糖尿病组相比,姜黄素能够改善血清和肝脏甘油三酯、总胆固醇、低密度脂蛋白、谷草谷丙转氨酶等生化指标(P<0.05)。研究发现,姜黄素降低糖尿病小鼠肝脏糖皮质激素水平(P<0.05),并伴随着糖皮质激素受体蛋白表达明显降低,显著性低于糖尿病模型组(1.17±0.23 vs Cur 100 mg/kg0.40±0.20,P<0.01)。病理切片确证姜黄素缓解糖尿病肝脏脂肪变性。结论:姜黄素具有调节糖尿病小鼠代谢紊乱的药理活性,可能是通过缓解糖皮质激素信号通路紊乱的作用。 Objective: To investigate the effects of curcumin on hepatic metabolic disorders in type 2 diabetic( T2DM)mice. Methods: T2 DM mice were established as previous method,and then were randomly divided into curcumin-treated group( 50 mg / kg,100 mg / kg) and vehicle-treated DM group. The normal mice were considered as control group.Through automatic analysis,enzyme linked immunosorbent assay,western blot and hematoxylin-eosin staining,the present study explained the pharmacological effects of curcumin. Results: Compared with diabetic mice,treatment with curcumin significantly decreased the levels of triglyceride,total cholesterol and low density lipoprotein in serum( P 〈0. 05) and inactivated the activities of glutamic-oxalacetic transaminase and glutamic-pyruvic transaminase in liver tissues( P 〈0. 05). Curcumin also reduced the expression of GR protein to relative 0. 40 ± 0. 20,which was less than relative 1. 17 ±0. 23 in diabetic mice( P 〈0. 01). Furthermore,histopathological analysis demonstrated that curcumin could improve the structural abnormalities in diabetic liver tissues. Conclusions: Curcumin has the ability of alleviating metabolic disorders in T2 DM mice,which may depend on effectively attenuating diabetes-induced glucocorticoid excess.
