榄香烯抑制p21基因表达逆转肺癌化疗耐药的实验研究 被引量:8

摘要 目的:研究榄香烯逆转肺癌化疗耐药的作用,及其对P21基因表达的影响。方法:体内研究:榄香烯、多柔比星及两者联合预处理小鼠5 d后,检测各组小鼠血中P21基因表达情况;复制Lewis肺癌小鼠模型,观察肿瘤直径长至1 cm所耐时间;然后分别予榄香烯、多柔比星及两者联合用药10 d,称瘤重,并检测肿瘤组织中P21基因表达情况。体外研究:采用榄香烯、多柔比星及二者联合预处理的小鼠血清培养A549肺癌细胞,48 h后检测各组细胞的增殖情况。结果:榄香烯联合多柔比星预处理组小鼠血清中P21基因表达水平与二者单用预处理组及正常组无统计学差异;二者联合预处理组小鼠血清培养A549肺腺癌细胞的增殖速度小于二者单用组;榄香烯联合多柔比星的抑瘤率明显高于二者单用,且二者联用组小鼠肿瘤组织中P21基因表达水平低于多柔比星组,但高于榄香烯组。结论:榄香烯具有逆转肺癌化疗耐药的作用,这一作用的可能机理与其抑制P21基因表达相关。 Objectives: To research the effect of Elemene reversing lung cancer cells' chemotherapy drug resistance and function of Elemene intervening p 2 1 gene expression. Methods : In vivo study : the p 2 1 gene expression of mice blood,which were pre-treated with Elemene,Doxorubicin and both them for five days,was detected. After duplicating mice model with Lewis lung cancer,we recorded how long tumor diameter grew from 0 cm to 1 cm. When tumor diameter was1 cm,the mice were treated by Elemene,Doxorubicin and both together,respectively. After ten days,the tumor qualities were weighted and p21 gene expressions of tumor were detected. In vitro study: the A549 lung cells were cultured with mice serum which were pre-treated by Elemene,Doxorubicin and both together,respectively. The cell proliferation was tested with Flow cytometry instrument after 48 hours. Results: The p21 gene expressions of mice serum had no statistical difference between groups. The proliferation rate of A549 lung cancer cell was higher in mice which were pre-treated with Elemene and Doxorubicin than in mice which were pre-treated with Elemene or Doxorubicin. Anti-tumor rate of Elemene and Doxorubicin was higher than Elemene or Doxorubicin. The p21 gene expression of tumor was lower in Elemene and Doxorubicin group than that in Doxorubicin group,but higher than that in Elemene group. Conclusions: It was reversed by Elemene that the lung cancer cell resisting chemotherapy drug and the mechanism may be related with Elemene inhibiting p21 gene expression.
