摘要
目的探讨Caspase12在糖尿病大鼠逼尿肌细胞内质网应激中的表达及作用。方法在不同的时间点(4、8、12、16周后诱导1型糖尿病大鼠模型),通过注射链脲菌素诱导糖尿病大鼠模型。在电镜下观察内质网形态,应用TUNEL技术检测糖尿病大鼠膀胱逼尿肌细胞的凋亡水平,采用RT-PCR技术测定逼尿肌细胞内Caspase12 mRNA的表达水平,采用Western-blot技术检测逼尿肌细胞内的Caspase12蛋白表达水平。结果电镜下内质网呈不规则改变,膜表面附着大量核糖体;逼尿肌细胞凋亡水平随着时间延长呈显著升高趋势;Caspase12的mRNA和蛋白水平曲线随病理过程的延长而增加。结论 Caspase12参与糖尿病大鼠逼尿肌平滑肌细胞的内质网应激。
Objective To explore the expression and effect of Caspase12 in the endoplasmic reticulum stress of diabetic rat detrusor muscle. Methods At different time points(4,8,12,16 weeks after induction of type 1 diabetic rat model),diabetic rat model was induced by streptozocin injection.Endoplasmic reticulums was observed under electron microscope,apoptosis level of diabetic rats′ detrusor smooth muscle cell was detected by using TUNEL technique,m RNA of Caspase12 was detected by using RT-PCR technique,protein expression of Caspase12 was detected by Western-blot technique. Results Irregular-shaped cavity changes in the endoplasmic reticulum was seen under the electron microscope,the membrane attached to a large number of ribosome,the apoptosis level of detrusor muscle cells showed a significant elevated trend with the extension of time,the levels of caspase12′mRNA and protein also indicated the increased curves with the prolonged pathological course. Conclusion Caspase12 is involved in the endoplasmic reticulum stress of the diabetic rat detrusor smooth muscle cell.
