肿瘤转移相关基因1在食管鳞癌中的表达及其与微血管生成的关系分析 被引量:3

摘要 目的探讨肿瘤转移相关基因-1(MTA1)在食管鳞癌(ESCC)中的表达及其与微血管生成的关系。方法选取本院胸心外科2013年3月-2014年1月的107例ESCC手术切除病例及56例正常食管组织的石蜡包埋组织样本作为研究对象,采用免疫组化检测MTA1在ESCC中的表达,应用CD34标记ESCC中的微血管内皮细胞,并计数微血管密度(MVD),分析MTA1基因表达与微血管生成之间的关系。结果 ESCC中的MTA1蛋白高表达率为50.4%,显著高于正常食管组织的23.2%,差异有统计学意义(P〈0.05)。ESCC中的MVD计数与正常食管组比较,差异有统计学意义(P〈0.05)。MTA1高表达率、MVD计数与ESCC的T分期、淋巴结转移显著相关,不同分组间比较,差异有统计学意义(P〈0.05)。MTA1的高表达率、MVD计数在ESCC不同TNM分期中比较,差异有统计学意义(P〈0.05)。在ESCC中,随着MTA1蛋白表达强度的增加,MVD均数亦逐渐增加,经单因素方差分析检验,差异有统计学意义(P〈0.05)。结论 MTA1高表达与ESCC的临床进展、微血管生成密切相关,可能成为潜在的抗食管鳞癌微血管生成的靶点。 Objective To explore the expression of metastasis associated gene 1(MTA1) in esophageal squamous carcinoma(ESCC) and the relationship between MTA1 and micro-angiogenesis. Methods 107 cases with paraffin embedding tissue samples of ESCC surgical removal and 56 cases of paraffin embedding tissues samples with normal esophageal tissue from March 2013 to January 2014 of cardiothoracic surgery in our hospital were selected as the research objects,the expression of MTA1 in ESCC was tested by using immunohistochemical.Microvascular endothelial cell in ESCC was marked by CD34.Density of MVD was counted.The relationship between the expression of MTA1 and micro-angiogenesis generation was analyzed. Results The high expression rate of MTA1 in ESCC was 50.4%,which was higher than 23.2% in the normal esophageal tissue,with significant difference(P〈0.05).There was significant difference in the count of MVD between ESCC and normal esophageal tissue(P 〈0.05).The high expression rate of MTA1 and count of MVD had significantly relationship with T periodization and lympho node metastasis of ESCC,and compared with different groups,the difference was statistics significance(P〈 0.05).Among the different TNM stage of ESCC,the high expression rate of MTA1 and the count of MVD had significant difference(P〈0.05).In ESCC,with the increased of MTA1 expression,the mean of MVD gradually increased,and by the single factor analysis of variance test,the difference was statistics significance(P〈0.05). Conclusion The high expression of MTA1 has closely relationship with clinical development and micro-angiogenesis generation of ESCC,and may becomes the potential target of esophageal squamous cell carcinomas microvascular generated.
