两种制备大鼠胚胎成纤维细胞的方法比较

摘要 目的比较酶消化法和组织块贴壁法对大鼠胚胎的皮肤成纤维细胞的培养情况,探讨何种方法能更加高效快捷地获取和培养大鼠的胚胎成纤维细胞。方法选择孕14 d大鼠胚胎,取其背部皮肤,分别采用酶消化法和组织块贴壁法获得原代的胚胎成纤维细胞,观察其生物学特点,进行传代、冻存、复苏和HE染色,并绘制细胞生长曲线。结果两种方法都能成功培养出胎鼠成纤维细胞,但组织块贴壁法相对酶消化法更便捷,不易污染,成活率高;传代和复苏后,两种方法获得的细胞无明显差异。结论组织块贴壁法是一种更适应胚鼠成纤维细胞的培养方法。 Objective To explore which way can be more efficient and faster to acquire and cultivate rat embryonic fibroblasts in this study by adopting enzyme digestion and tissue explants adherent method to compare cultivation of embryonic fibroblasts from rat′s skin. Methods Embryos of pregnancy rats(14 days) were taken to get its dorsal skin.Enzyme digestion and tissue explants adherent method were used respectively to obtain the primary rat embryonic fibroblasts and then to observe its biological characteristics,which was passaged,cryopreserved,resuscitated and done with HE staining.And growth curve of cells was drawn. Results Rat embryonic fibroblasts were successfully cultivated by both methods.Compared with the enzyme digestion,the tissue explants adherent method was more convenient,not easily polluted and with high survival rate.There was no difference between cells acquired by the two methods after being passaged and resuscitated. Conclusion Tissue explants adherent method is a better way to cultivate rat embryo fibroblast.
