Simultaneous Determination of Ferulic Acid and Gastrodin in Naoluoxintong Capsule by Dual-wavelength High-performance Liquid Chromatography-Diode Array Detector

摘要 目的 建立双波长同时测定脑络欣通胶囊中天麻素和阿魏酸含量的方法,为脑络欣通的质量控制提供依据。方法 采用高效液相色谱-二极管阵列检测器和Welch Materials C8色谱柱(250mm×4.6mm,5μm)。流动相:乙腈-0.01%磷酸;梯度洗脱(乙腈:0-10min,6%;10-30min,6%→35%);流速:1.0mL/min;柱温:35℃;检测波长:320nm(阿魏酸)和222nm(天麻素)。结果 在30min内,实现了阿魏酸与天麻素双波长同时测定,二者含量分别在0.20-2.39μg(r=0.999 9)和0.20-2.41μg(r=0.999 8)范围内与峰面积呈现良好的线性关系,平均加样回收率分别为99.5%和99.1%(n=6)。结论 双波长检测对复方中阿魏酸与天麻素定量更准确,二者同时测定能有效提高分析效率,该方法可用于脑络欣通胶囊的质量控制。 Objective To establish a dual-wavelength method for simultaneous determination of the content of gastrodin and ferulic acid in Naoluoxintong Capsule, and to provide a basis for the quality control of Naoluoxintong Capsule. Methods High-performance liquid chromatography-diode array detector and Welch Material C8 column (250 mm × 4.6 mm, 5 μm) were applied. The conditions for detection were a mobile phase of acetonitrile and 0.01% phosphoric acid, gradient elution (acetonitrile: 0-10 min, 6%; 10-30 min, 6%-35%), a flow rate of 1.0 mL/min, a column temperature of 35 ℃, and a detection wavelength of 320 nm (ferulic acid) or 222 nm (gastrodin). Results 〖JP2〗The dual-wavelength simultaneous determination of the content of ferulic acid and gastrodin was realized within 30 min. The content of ferulic acid and gastrodin was in a good linear relationship with peak area within 0.20-2.39 μg (r=0.999 9) and 0.20-2.41 μg (r=0.999 8), respectively. The average recovery rates of ferulic acid and gastrodin were 99.5% and 99.1%, respectively (n=6). Conclusion The dual-wavelength determination of the content of ferulic acid and gastrodin in the compound is more accurate. Simultaneous determination of the two ingredients can increase analysis efficiency effectively, and can be applied as the method of quality control for Naoluoxintong Capsule.
